Objective The main purpose of the test is to investigate the effects of PIRES-egr-1-IFN-γ-Endostatin limpsome combined irradiation to the tumor in vivo, even the depressive effects of endostatin on angiogenesis and immniolgy effects of IFN-γto tumor . It will provide scientific theories and experience basements to research the genetherapy in clinic.Methods To the limipsome of PIRES-egr-1-IFN-γ-Endostatin ,and inject the limipsomes into the tumors of heptic-carcinoma in rats , then give local irradiation to the mass. The limipsome will be induced to express and the growth of the mass will be checked, and the diameter of the mass will be getted . Then the expression levels of IFN-γand Endostatin in mouse serum with different vector dosage and different time was measured by ELISA. We can biopsy the mass and stain in H&E and ISH . The number of vascular in H&E staining and the expression of CD31 in ISH staining were counted.Results①The growth of the mass was inhibited obviously in a dose dependent and a time dependent manner after injection of plasmid and radiating, and it was statistically significant obviously that tumor growth in radiation group and plasmid +radiation group was slower than which in control groups (p<0.01).②Radiation can induce the expression of INF-γand Endostatin in a dose dependent manner and its level increased markedly and reached to a peak at the 3td week. The expressions of INF-γcan maintain steadily for about 4~5 weeks.③The number of vascular in H&E staining and the expression of CD31 in ISH staining were different in diferent groups (P<0.01) .Conclusions Radiation can induce the expression of IFN-γand Endostatin gene in a dose-dependent and atime-dependent manner. Genetherapy of PIRES-egr-1-IFN-γ-Endostatin combined with irradiation could enhance the antitumor effection more effective than any one of genethreapy and radiotherapy only.
