Sodium alginate is one kind of natural linear polysaccharide which obtains from the brown alga.As a natural polymer material, alginate has a good biocompatibility, abundant raw materials, accessible and inexpensive and widely used in tissue engineering and drug release, and other fields. The paper take the sea alginic acid sodium as a raw material, the thesis mainly includes the following three parts:Alginate was hydrophobically modified through oxidation of the alginate with sodium periodate, followed by reaction of the oxidized alginate and dodecylamine. The modified alginate was characterized with IR, 1H-NMR. The viscosity and Fluorescence spectra of the polymer solution were measured. The modified alginate was made into microsphere beads in aqueous solution of sodium chloride and calcium chloride. The beads were used to load Buloh phenol. The in vitro controlled release was investigated. It was usually considered that alginate gels used in tissue engineering degradate very slowly and the degradation products molecular are very great,so it is not easy to discharge from vitro, therefore, its application receives certain limit, based on oxidized alginate and take the calcium chloride as the crosslinking agent, the preparation of calcium ion crossing linking oxidized alginate hydrogels maintains both mechanical properties and the good degeneration performance.Take the ethylenediamine as the crosslinking agent to prepare alginate covalently cross-linked hydrogels and studied the performance influencing factor of the covalently cross-linked hydrogels and made the performance contrast with the ion crossing linking gelatin ,which made a good theoretical guidance for more reasonable use of alginate hydrogels in tissue engineering, drug delivery systems and three-dimensional cell culture systems.
