Objective: To investigate the relationship between aldehyde dehydrogenase-2 (ALDH2) gene polymorphisms and the efficacy of nitroglycerin (GTN) in the CHD patients with stable angina pectoris.Methods: One hundred and thirteen CHD patients with stable angina pectoris, who were confirmed by coronary angiography (one of the main coronary arteries narrows≥50%) and only sublingual nitroglycerin during acute stable angina pectoris attack were involved in this study. The whole patients were divided into two groups according to the efficacy of only sublingual nitroglycerin during acute stable angina pectoris attack: efficacious group (Group A) and inefficacious group (Group B). Among Group A, according to the time which was cost by relief of angina pectoris after sublingual nitroglycerin, we divided the patients into Group A1 (≤5 min, n=69) and Group A2 (>5 min, n=18). We obtained the DNA from 2ml peripheral blood of each patient. Polymorphism chain reaction (PCR) and reverse direct sequencing were performed to screen the polymorphism of ALDH2 gene in exon 12.Results: There is a Glu to Lys polymorphism of ALDH2 gene in exon 12 at position 504 (Gly504Lys). The frequencies of G and A allele in the total population was 0.819 and 0. 181, respectively. No significant difference was found in the genotype distribution of ALDH2 gene between Group A and Group B (G/G: 74.713% vs. 61.538%, P>0.05; A/A: 6.897% vs. 11.538 %, P>0. 05). Among Group A, we did not find the significant difference in the distribution of ALDH2 gene between Group A1 and Group A2 (G/G: 73.913% vs. 77.778%, P>0.05; A/A: 4.348% vs. 16.667%, P>0.05), either. And ALDH2 genotype also did not show any significant difference between Group A1 and Group B (G/G: 73.913% vs. 61.538%; P>0.05; A/A: 4.348% vs. 11.538%, P>0.05).Conclusion: There is a Glu to Lys polymorphism of ALDH2 gene in exon 12at position 504 (Gly504Lys). The genotype of G/A polymorphism in ALDH2gene is not associated with the efficacy of nitroglycerin (GTN) in theCHD patients with stable angina pectoris.
