Objective:Made the acute epilepsy model by Lithium-Pilocarpine after preconditioning with low-frequency rTMS,observed the epileptic attack behavior and the expression of Bcl-2/Fas and Caspase-3 genes in the hippocampal neurons of rats.We objectived to explicit the anti-epilepsy function and the possible mechanism of rTMS,and to provide certain proofs for the study and development of epilepsy.Methods:Rats were randomly divided into five groups by the frequency(0 Hz,0.3Hz,0.5Hz,0.8Hz and 1.0Hz stimulation groups).Making the acute epilepsy model by Lithium-Pilocarpine after pretreatment,observed the latent period and the performance of epileptic seizures within 70 minutes after intraperitoneal injection of Pilocarpine.21 adult male Wistar rats were randomly divided into three groups:the blank control group were 6 rats;the sham stimulation group were 9 rats;the 0.5Hz stimulation group were 6 rats.Seizure was induced by hithium-Pilocarpine in the sham stimulation group and the 0.5Hz stimulation group after pretreatment. Then the two groups were executed at 12 hours,24 hours,72 hours after seizure.The brain tissues were studied histologically with HE staining to observe the neuronalalterations microscopically,and immunohistochemistry study was carried out for the observation of the expressions of Bcl-2/Fas and Caspase-3 genes.Results:The latency period each of 0.3Hz,0.5Hz,0.8Hz and 1.0Hz stimulation groups was prolonger than model group(P<0.05 or P<0.01).The latency period in the 0.5Hz stimulation group was prolonger than 0.3Hz,0.8Hz and 1.0Hz stimulation groups(P<0.05).The times of jump each of 0.3Hz,0.5Hz,0.8Hz and 1.0Hz stimulation groups were less than the model group respectively(P<0.05 or P<0.01).The times of epileptic attack behavior in 0.5Hz stimulation group was less than 0.3Hz,0.5Hz,0.8Hz and 1.0Hz stimulation groups(P>0.05).The epileptic seizure of rats didn't increase by rTMS(1.0Hz,75%MT).Appropriate transcranial magnetic stimulation is safe.The expression of Bcl-2 genes in the hippocampns neurons CA1 in the 0.5Hz stimulation group were more than the sham stimulation group,at 12h(P<0.05),24h(P<0.01),72h(P>0.05).The expression of Fas genes in the hippocampns neurons CA1 in the 0.5Hz stimulation group were less than the sham stimulation group,at 12h(P<0.01),24h(P<0.01),72h(P>0.05).The expression of Caspase-3 genes in the hippocampns neurons CA1 in the 0.5Hz stimulation group were less than the sham stimulation group,at 12h(P<0.05),24h(P<0.05),72h(P<0.05).Conclusion:1 Appropriate transcranial magnetic stimulation could prolong the latent period of epilepsy seizures,diminish the symptom and level of epilepsy attack,and have conspicuous anti-epilepsy effect. 2 Appropriate transcraniat magnetic stimulation has protective function through regulating the expression of Bcl-2/Fas and Caspase-3 genes probably.
