Objective: To observe the effects of Ligustrazine combined with Benazepil on the macrophage infiltration and expression of Tumour necrosis factor-α(TNF-α) in the renal interstitium of Goto-Kakizaki wister rates (GK rats) , and explore the possible function of renoprotection and mechanism of Ligustrazine combined with Benazepil in the earlystage of DM.Methods: 40 male GK rats were randomly divided into model group (n=10), Benazepril group (n=10), TMP group (n=10), TMP+ Benazepril group (n=10), take another Wistar rats as normal group (n=10). The model and normal group were respectively treat with physiological saline 5ml·kg-1·d-1 and the other were respectively treat with Benazepril 1.6mg·kg-1·d-1 TMP 150mg·kg-1·d-1,TMP 150mg·kg-1·d-1+ Benazepril 1.6mg·kg-1·d-1intragastrically for 16 weeks. The 24hr urinary protein count were determined at the 8th, 12th, 16th week. At the end of 16th week, killed all of the rats, gathered blood from abdominal aorta immediately, and measured their serum creatinine (SCr),blood urea nitrogen (BUN),triglyceride (TG),total cholesterol (TC), kidneys were harvested and the histological changes of the kidneys were observed after HE satin. The expression of TNF-αand ED-1 in renal interstitium was detected by immunohistochemistry.Results: Compared with the normale group, the TG,TC and 24hr urinary protein count of model group increased significantly (p<0.01), the expression of TNF-α,ED-1 in the renal interstitium of diabetic rats increased significantly (p<0.01); Compared with the model group, the 24hr rurinary proteine count,the content of serum creatinine and blood urea nitrogen were decreased to a diffrent degree (p<0.05或p<0.01), the TG,TC , the expression of TNF-αand ED-1 in the renal interstitium of each treat group were decreased to a diffrent degree (p<0.05), and the TMP + Benazepril treated group were significantly decreased.Conclusions: Ligustrazine combined with benazepil can improve renal function and reduce interstitial pathological changes of GK rats. The mechanism may be related to decreasing excretion of proteinuria, downregulating macrophages infiltration and the expression of TNF-αin renal interstitium.
