| Objective: To optimize the methods of building models of experimental type 2 diabetes and establish the optimal technique. To investigate the pharmacological influence of Chinese medicine Jiangtang Capsule on type 2 diabetic rat models. Methods: Wistar rats were administered and feed in difference styles to induce experimental type 2 diabetic animal model, results were evaluated by successful rate of diabetic rat model, stabilization time?operation means and the optimal technique was established. Stabilization time of blood sugar and changes of body weight in animal models confirmed this animal model and the optimal technique. The pharmacological influences of Jiangtang Capsule on type 2 diabetic animal model were investigated. The changes of blood sugar, body weight, insulin release, homeostatic model of insulin resistance (Homa-IRI) calculations and lipid efficiency in experimental diabetic rats were monitored after the treatment of Jiangtang Capsule. Results: Blood sugar in animal models of experimental type 2 diabetes stabilized in established period. Before Jiangtang Capsule or positive samples administered, all STZ groups had higher glucose concentrations to control group, and significantly higher triglyceride (TG), total cholesterol (TC), low-density lipoprotein (LDL-C). Jiangtang Capsule or positive samples, administered for two months reduced TG (P<0.05), TC (P<0.05), LDL-C (P<0.05) and increased HDL-C (P<0.05). Compared with control group, Met groups had a somewhat lower glucose. Jiangtang capsule and positive samples had significantly higher insulin sensitivity index (ISI) (P<0.01) to control group. Conclusion: The optimal technique was available. Animal model has character of type 2 diabetes. The study suggested that Jiangtang capsule can improve the hyperlipidemia, the functional cells reverse and insulin resistance in fat-fed, streptozotocin-induced diabetic rats.
