Object: To investigate the expression of Protein kinase CK2 and its relationship with the development, progress, invasion and metastasis of squamous cell carcinoma of larynx (LSCC).Method: Immunohistochemical SP staining method was used to assess the expression of CK2 in 18 cases of normal laryngeal mucosa, 14 cases of polyp of vocal cord, 11 cases of larynx papilloma and 50 cases of LSCC patients. And RT-PCR was used to detect the expression of CK2αmRNA and CK2βmRNA in 50 cases of LSCC patients. The relationship between CK2αand CK2βwas evaluated.Result: The lowest rate of positive expression of CK2αand CK2βwas found in normal laryngeal mucosa, polyp of vocal cord cases, tissues close to carcinoma by 1.0cm, nonmetastatic lymph nodes (11.11%,21.43%,20.00%,10.53% and 11.11%,14.29%,15.00%,15.79% respectively). The expression rate of CK2αand CK2βinvolved in tissues of laryngeal carcinoma and metastatic lymph nodes of neck was the highest (92.00%,84.21% and 88.00%,78.95% respectively) compared with that of laryngeal papilloma and tissues close to carcinoma by 0.5cm (54.55%,55.00% and 45.45%,50.00% respectively). In the group of LSCC, the expression of CK2αin G2+G3 was higher than that in G1 (p=0.033). While the age of the patients, TNM stage and lymphatic metastasis didn't change the expression of CK2αobviously (p=0.435,p=0.173,p=0.190). The expression of CK2βcorrelates to the differentiation grading and lymphatic metastasis in LSCC patients: CK2βin G2+G3 was higher than that in G1 (p=0.048); the expression of CK2βprotein in LSCC with the metastasis of lymph nodes was higher than that in LSCC without metastasis (p=0.018), but not to the age and TNM stage(p=0.300,p=0.204). The result from RT-PCR was highly consistent with that from immunohistochemical SP staining. The expression of CK2αin LSCC patients shows positive correlation to that of CK2β(r=0.735,p=0.001).Conclusion: The over expression of protein kinase CK2 maybe an accelerator to the formation and development of LSCC. Protein kinase CK2 may be one of the predictors for the malignant grade of LSCC. To inhibit the over expression might be new therapeutic methods for LSCC.
