Objective: Analysis of the CK8protein expression and its clinical pathologicalsignificance in laryngeal squamous cell carcinoma.Methods: Selected78cases of laryngeal squamous cell carcinoma, in which the56cases without lymph node metastasis(included16cases of highly differentiated,18cases of middle differentiation and22cases of poorly differentiated),22cases withlymph node metastasis;10cases of laryngeal papilloma,12cases of laryngeal polypsand16cases of normal controls(taken the mucosa away from mass>2cm as normallaryngeal mucosa in laryngeal surgical specimens), phase I and II are42cases, phaseIII and IV are36cases; the expression levels of CK8mRNA were detected byRT-PCR, the expression of CK8protein were detected by immunohistochemistry,analysis of the relationships among the expression of CK8gene, pathological gradeand lymph node metastasis.Result: RT-PCR results showed that the expression of CK8mRNA in highdifferentiation carcinoma was0.96±0.42, middle differentiation carcinoma was1.71±0.38, poorly differentiated carcinoma was1.85±0.51, laryngeal squamous cellcarcinoma with lymph node metastasis group was2.25±0.45, papilloma was0.86±0.29, polyps was0.23±0.08, normal laryngeal mucosa was0.21±0.07; thestatistical analysis showed that the expression level of CK8mRNA associated withthe degree of differentiation and lymph node metastasis, the P values were0.048and0.032, respectively. Immunohistochemistry showed that the positive expression ratewas46.4%(26/56) in without metastasis laryngeal carcinoma group,12.5%(2/16) inhigh differentiation carcinoma,55.5%(10/18) in middle differentiation carcinoma,63.6%(14/22) in poorly differentiated carcinoma,81.8%(18/22) in laryngealsquamous cell carcinoma with lymph node metastasis,10%(1/10) in papilloma, but the normal laryngeal mucosa and throat polyps were negative. The statistical analysisshowed that the differences of positive expression rate have significant(p<0.01)between normal mucosa and carcinomas, papillomas and carcinomas, the differences of positiveexpression rate were significant(p<0.01) between without lymph node metastatic carcinoma andlymph node metastasis carcinoma, while the difference were no significant(p>0.05)betweennormal mucosa and papilloma.Conclusion:1.CK8gene is highly expressed in laryngeal carcinoma.2.The positive expression of CK8gene associated with the degree of differentiationand lymph node metastasis, with the degree of differentiation decreasing, theexpression increasing, the lymph node metastasis in laryngeal squamous cellcarcinoma positive expression rate of CK8gene higher than non-laryngeal squamouscell carcinoma metastasis.3. CK8gene may be closely related with the degree of laryngeal cancer, and maypromote lymph node metastasis in laryngeal cancer at the same time. CK8geneaccelerates the occurrence and development of laryngeal cancer.
