Objective:To investigate the significance on relationship between Ubiquitin hydrolase protein gene product PGP9.5 and COX-2 protein expression in human colorectal carcinoma,Method: colorectal specimens from 68 patients with esophageal carcinoma and 8 normal subjects were randomly collected and studied, colorectal carcinoma species were examined by polymerase chain reaction (PCR) using PGP9.5-specific primers, A search for COX-2 that is specific for colorectal carcinoma was also carried out by PCR. The immunohistochemistry staining for PGP9.5 and COX-2 protein was performed.Results: the normal colorectal squamous epithelium showed PGP9.5 and COX-2 negative expression. PGP9.5 was expressed in 56 of tumors, mainly in cytoplasm and membrane of tumor cell, weakly in stromal cells. The expression of PGP9.5 was associated with lymphnode metastasis(P<0.05), clinical stage (P<0.05), and infiltration depth(P<0.05), but was not associated with gender, age, pathological classification and histological grade(P>0.05). COX-2was expressed in 52 of tumors, mainly in membrane of tumor cell. The expression of COX-2 was associated with lymphnode metastasis(P<0.05), infiltration depth(P<0.05), but was not associated with gender, age, pathological classification, clinical stage and histological grade(P>0.05). The expression of PGP9.5 and COX-2 show the positive correlation,kaplan-Meier method shows a significantly lower survival probability for patients with PGP9.5 positive comparing to patients with COX-2 positive (P<0.01),and PGP9.5 was an independent prognostic factor for survival of patients with colorectal cancer.Conclusion: PGP9.5 are found in human colorectal carcinoma tissues widely which shows PGP9.5 expression may play a considerable role in the progression of colorectal carcinoma and served as an independent factor of prognosis. Routine detection PGP9.5 expression may be useful for the prediction of treatment and prognosis in colorectal carcinoma. COX-2 protein expression increases markedly in PGP9.5-positive carcinoma tissues, which might be one of mechanisms of signaling pathway in generation and development of human colorectal carcinoma.
