Background:Obstructive sleep apnea/hypoventilation syndrome (OSAHS) is a common disease, associated with nocturnal intermittent hypoxia. Researches show that OSAHS is assumed to play a part in hypertension and vascular dysfunction. Hypoxia-inducible factor 1 (HIF-1) is a transcription factor that functions as a master regulator of oxygen homeostasis, transcriptional activity of which depend on the amount of HIF-1α. Plasminogen activator inhibitor-1(PAI-1) is one of the hypoxia-inducible genes directly regulated by HIF-1α, and link to hypertension and atherosclerotic diseases. Shenmai has protective effect on heart and blood vessel in ischemia-reperfusion, which is similar with chronic intermittent hypoxia (CIH) pathologically and physiologically. The mechanism of OSAHS leading to cardiovascular is unclear. In this study, we establish animal model, to investigate the effect on heart and vessel of CIH, and the intervention of shenmai injection.Objective:To investigate the influence on HIF-1αand PAI-1 in cardiovascular caused by CIH in OSAHS, and the effect of shenmai injection on plasminogen system.Methods:Thirty male mice were divided into three groups: an air control group, a CIH group and a shenmai injection group. Immunohistochemistry was used to examine the expression of HIF-1αand PAI-1 in the myocardial cells and vascular. RT-PCR was used to measure PAI-1mRNA.Results:The expression of HIF-1αand PAI-1 in myocardial cells and vascular of the CIH group significantly increased compared with that of the air control group and that of the shenmai injection group. The expression of HIF-1αand PAI-1 in myocardial cells and vascular of the air control group was not significantly different from that of the shenmai injection group.Conclusions:CIH enhances the expression of HIF-1αand target gene PAI-1 in myocardial cells and vascular. Shenmai injection decreases the expression of them.
