Objective: To explore anti-rejection effect of PLA-O-CMC nanoparticles in hepatocyte xenotransplantation.Methods: The isolated rat hepatocytes of the concentration 5×105cells/ml were inoculated into the RPMI1640 medium and divided into 3 groups: group I, group II and III. Group I was co-cultured with PLA-O-CMC nanoparticles. The cells were incubated in 37℃5%CO2 incubator with intermittent vibrating. Afrer 12h, group I and II were co-cultrued with type I collagen gel. 12h later, 5ml hepatocytes co-cultured with PLA-O-CMC nanoparticles embedded in collagen gel(Group I), 5ml hepatocytes emdbedded in collagen gel(Group II), and 5ml pig free hepatocytes suspension(5.0×107cells) (group III) were transplanted into abdominal cavity of SD rats at 48h after D- gal introperitonear injection (1.2g/kg). The concentrations of IL-2, IFN-γ, IgG, and IgM in serum of recepators were evaluated. The transplanted hepatocytes were observed by optical microscope.Results: There were no significant difference of serum IL-2 and IFN-γbetween the 4 groups. The concentration of serum IgG went up after operation in each group, reached the pinnacle 3d after operation, and began to decline in the following days. Compared with group II, there was significant difference between group I and II, so did between group I and III. The concentration of serum IgM of each group was lower than which of the ALF rat model, and the group I was higher than group II and III 1d after operation. 48h later, there were no significant differences of each group compared with the first day. After 72h, the serum IgM of each group began to rise, and the group I was more obvious than the rest group. The transplanted hepatocytes survival time of group I were longer than that of other groups.Conclusion: The PLA-O-CMC nanoparticles can protect the heaptocytes which intraperitoneal transplanted for treating ALF rat from immunologic rejection in some degree.
