In the present work HPLC-UV and LC-MS/MS methods were used for determination of Emodin in rat tissues after i.g administration of Polygonum cuspidatum Sieb.et Zucc.extract.In this thesis the study on determination of bioactive compound of Polygonum cuspidatum Sieb.et Zucc.is to provide scientific evidence for how its bioactive compound work.Six reliable HPLC-UV methods are developed to determine Emodin in rat kidney,lung,liver,spleen,heart and intestine,respectively,The internal standard(IS) is 1,8-dihydroxyanthraquinone,and A LC-MS/MS method is developed to determine emodin in rat brain.Sample preparation is achieved by liquid-liquid extraction.The regression equation of Emodin in the rat kidney was y=0.2629x-0.0512, and the correlation coefficient was 0.9995.The linear range of the calibration curve were 0.30~19.10μg/g,and the limit of detection was 0.12μg/g.The extraction recovery was 91.7~101.6%.The concentrations of emodin in rat kidneys were measured.The Cmax was 4.19 pg/g occurring at 30min.The regression equation of Emodin in the rat lung was y=0.3247x-0.0808, and the correlation coefficient was 0.9997.The linear range of the calibration curve were 0.33~27.50μg/g,and the limit of detection was 0.12μg/g.The extraction recovery was 97.0~102.9%.The concentrations of emodin in rat lungs were measured.The CmaxWaS 20.83μg/g occurring at 10min.The regression equation of Emodin in the rat liver was y=0.1071x-0.0039, and the correlation coefficient was 0.9995.The linear range of the calibration curve were 0.47~39.22μg/g,and the limit of detection was 0.19μg/g.The extraction recovery was 84.4~101.2%.The concentrations of emodin in rat livers were measured.The CmaxWaS 9.65μg/g occurring at 10min.The regression equation of Emodin in the rat spleen was y=0.3317x-0.0974, and the correlation coefficient was 0.9935.The linear range of the calibration curve were 0.33~5.50μg/g,and the limit of detection was 0.18μg/g.The extraction recovery was 91.3~98.6%.The concentrations of emodin in rat spllens were measured.The Cmax was 1.08μg/g occurring at 15min.The regression equation of Emodin in the rat heart was y=0.5298x+0.0271, and the correlation coefficient was 0.9971.The linear range of the calibration curve were 0.11~2.20μg/g,and the limit of detection was 0.06μg/g.The extraction recovery was 88.9~102.0%.The concentrations of emodin in rat hearts were measured.The CmaxWaS 1.04μg/g occurring at 10min.The regression equation of Emodin in the rat intestine was y=0.2422x+0.0871,and the correlation coefficient was 0.9999.The linear range of the calibration curve were 0.60~358.50μg/g,and the limit of detection was 0.18μg/g.The extraction recovery was 92.8~97.2%.The concentrations of emodin in rat intestines were measured.The CmaxWaS 142.18μg/g occurring at 10min in small intestine.The Cmax was 99.58μg/g occurring at 12h in large intestine.The CmaxWaS 19.56μg/g occurring at 7min in cecum.The regression equation of Emodin in the rat brain was y=137.68x-314.62, and the correlation coefficient was 0.9999.The linear range of the calibration curve were 5.0~1000.0 ng/g,and the limit of detection was 1.50 ng/g.The extraction recovery was 94.1~7.2%.The concentrations of emodin in rat brains were measured.The Cmax was 808.50ng/g occurring at 10min.The results show that Emodin in Polygonum cuspidatum Sieb.et Zucc.extract distributs over rat tissues quickly,and is disposed of completely throughout 24h except intestine.The emodin concentration of intestine is the highest in these tissues,the concentration from high to low in tissues:intestine,lung,liver,kidney,spleen,heart,brain.The distributions of tissues correspond with its traditional therapeutic effect.
