BACKAROUND: The extrinsic processes, known as photoaging, is mainly resulted from ultraviolet radiation Recently, substantial progress has been made in understanding cellular and molecular mechanisms that bring about photoaging. But , there are still many questions on the fundamental molecular pathways. In addition, only few researches about Chinese people skin photoaging have been carried out, so It's necessary to investigate the cellular and molecular mechanisms of Chinese people skin photoaging with a new microarray technique.OBJECTIVE: To clarify further the genetic molecular mechanisms underlying the development of Chinese people skin photoaging to provide new clue for the elucidating of pathogenesis and obtain new evidences for the prevention and cure of skin photoaging.METHODS: Six pairs of skin specimens were obtained from sun-exposed (anterior ear skin) and sun-protected normal skin (post ear skin). Divided all the specimens into 2 groups: group1 (anterior ear skin) and group 2 (post ear skin). DNA microarray technique was applied for gene expression profiling analysis to compare the mRNA expression difference between the sun-exposed and sun-protected normal skin. Try to find the internal relationship between differentially expressed gene with GO,KEGG and NCBI/GENE data base.RESULTS: There are 172 differentially expressed genes in Beadarray chip, in which 99 up-regulating gens and 73 down-regulating genes. After primary classification with GeneBank research, there are 118 functionally classified genes, 7 unknown gene expression sequences related to signal transduction, regulation of transcription, immune response etc.CONCLUSION: We have verified the feasibility of microarray in the study of skin photoaging. we have found a lot of differentially expressed genes , which related to versatile cell function, and indicate that many kinds of genetic molecular mechanisms are associated with skin photoaging. There are some unclear relationships between soft tissue augmentation and differentially expressed genes in skin photoaging.
