Objective: To screen the related gene expression in gastric muscularis propria tumors and their surrounding tissues by gene chip assay,search for significantly differential expression gene,and discussion Preliminarily on gene expression changes and the possible bioinformatics meaning of gastric stromal tumors and the tumor to the muscularis propria week organization,and to guide the accurate diagnosis and targeted therapygastric of gastric stromal tumors originated form the muscularis propria.Methods: 6 cases of samples with gastric muscularis propria mesenchymal tumor and peritumoral tissue were treated by endoscopic treatment and laparoscopic surgery in Yantai Yuhuangding Hospital Affiliated to Qingdao University,and which are confirmed as gastric stromal tumors by the pathological examination and immunohistochemical examination.Before operation,all the patients were examined by the small probe ultrasonography,which showed that the lesions originated from the muscularis propria of the stomach.Then we can collect gastric muscularis propria mesenchymal tumor and peritumoral tissue during operation,and put each specimen immediately in liquid nitrogen to frozen preservation.cDNA was synthesized by reverse transcription with total RNA,then by in vitro transcription synthesised of biotin labeling cRNA.Then the fragment cRNA by purified and fragment was prepared into a suitable reaction system and hybridizationed with the Affymetrix mRNA expression profile chip.Different genes were screened according to the method of difference multiple.screened differentially expressed genes with bioinformatics analysis using PANTHER/KEGG database and Gene Ontology(GO)software in order to screen out the gene function classes and related signal pathways involved in tumorigenesis and development.Results: 1.3293 differentially expressed genes were detected in 6 cases of Gastric stromal tumor tissue,including 2588 up-regulated,and 705 down-regulated genes,especially in DPP10,ETV1,DKK4,CXCL14,MT1 M gene expression significantly.2.The function of these differentially expressed genes can be divided into many biological processes,such as cell differentiation,cell proliferation regulation,cell adhesion,cell signal transduction,cytoskeleton structure and so on.And these genes were involved in multiple signaling pathways related to tumor,such as Wnt pathway,Rap1 pathway,ECM receptor interaction,Ras signaling pathway,P13K-AKT signaling pathway,and cell adhesion.Conclusion: There are significant differences of gene expression between gastric stromal tumor and peritumoral tissue,the aitered expression of DPP10,ETV1,DKK4,CXCL14,and MT1 M might be involved in the regulation of gastric stromal tumor.These differentially expressed genes may provide an important biological basis and reference for the study of new molecular markers or therapeutic targets of gastric muscularis propria mesenchymal tumor.To conduct in-depth study of these genes and related regulation pathways,will contribute to early diagnosis and treatment of gastric stromal tumors,and to improve the long-term survival rate and quality of life of patients with gastric stromal tumors.
