The technology of screening bioactive compounds from traditional Chinese medicine is a hot sport in the field of pharmacy.Based on the reported screening method,Combining the specificity of receptor with the high efficiency of chromatography,we constituted an adrenergic receptor biochromatographic model and investigated its application in screening bioactive compounds of traditional Chinese medicine.The main work of the thesis includes the following three parts:1,β2-adrenergic receptor(β2-AR) were obtained by the tissues extraction and purified by affinity chromatography.Then,the receptors were immobilized on the surface of macroporous silica gel by mild chemical coupling method.The retention characteristic of theβ2-AR biochromatography was investigated with the several druds,such as salbutamol,propranolol, phentolamine et al.The results showed that theβ2-AR biochromatography model could retained receptor bioactive efficiently and had the good stability and specificity,which can be applied in investigating the interaction between receptor and drug,and screening bioactive compounds for the complex matrix such as traditional Chinese medicine.2,Angelicae sinensi,Scutellaria Baicalensis and Tinglizi were screened byβ2-AR biochromatographic model,and nine bioactive compounds were found.The results showed that receptor biochromatography can effectively screen the bioactive compounds,and have a potential application in screening bioactive compounds from traditional Chinese medicine.3,The association constant and the binding sites of immobilizedβ2-AR with palmatine and jatrorrhizine were determined by frontal analysis.The results showed that palmatine has one kind of binding sites,the association constant of palmatine for two kinds of models withβ2-AR is 2.407×104 M-1 and 4.302×104 M-1.Jatrorrhizine has two kinds of binding sites withβ2-AR,and the association constants are 1.095×105 M-1 and 3.016×102 M-1, respectively.
