ObjectiveTo investigate the in vitro histocompatibility of nano-CS/COL;then explore a new method to repair the bone defect with tissue engineering bone.MethodsNano-CS/COL scaffold was prepared by polyelectrolyte confocal laser-scanning microscopy.The well-grown cells of the third passage were co-cultured in vitro on the nano-CS/COL scaffold.Taking simple nano-CS/COL scaffold material as control,the histocompatibility of scaffold material and cells were comprehensively evaluated by cell adherence rate,growth curve,cell activity and cycle,and scanning electron microscope observation.1.BMSCs were isolated from SD rats and cultured.2.Cell surface antigen was detected by loss cell analytical method.3.nano-CS-TPP scaffold was made and co-cultured with BMSCs,observe the compatibility of cell and scaffold through calculating cell adherence ratio,drawing growth curve,detecting cell livingness and cell cycle,dyeing I collagen, observing cell by scan electron microscope.4.An bone defect(5mm in length) was created at two tibial in each mice.The composite grafts were implanted into segmental tibial defects in mice through open operation.The curative effect was evaluated by general observation, radiographic examination,histologic analysis at 6,12 week.Result1.The BMSCs could be obtained and cultured expressing CD29,CD44,CD105, CD106,not CD34,CD45.2.The interstice ratio and diameter of the nano-CS-COL scaffold is 85%~90%and 150μm,collagen can be saw.3.The BMSCs grow well and nano-CS-COL scaffold has good histocompatibility to BMSCs by scan electron microscope observation calculation et al. 4.In experimental groups,the bone defects can be repaired completely,much better than other two groups by general looking,X-ray and histologic analysis.Conclusion1.Nano-CS/COL scaffold can be used as tissue engineering biomaterials for bone marrow MSCs can well grow on it.2.the composite grafts of Nano-CS-COL/BMSCs quickly promote the new bone formation with better curative effect of tibial defects than other groups,it is a new scaffold in the bone engineering tissue.
