Objective To explore the potential pathogenic role of cell apoptosis,bcl-2,c-myc and proliferating cell nuclear antigen(PCNA) abnormality expression in the in the oncogenesis and development of gastric carcinoma.Methods The apoptotic index(AI) was tested by terminal deoxynucleotidyl transferase (TdT)-mediated dUTP-biotin nick end labeling(TUNEL) and the expression of bcl-2,c-myc and PCNA protein were tested by immunohistochemistry in 58 gastric carcinomas and corresponding para-carcinoma tissues.Results①The expression of bcl-2 protein was detectded in 29 of 58(50.00%) gastric carcinoma tissues and in 17(29.31%) of 58 corresponding para-carcinoma tissues.There was significant difference between the two groups(x~2=4.3214,P= 0.0376).②The expression of c-myc protein was detectded in 33 of 58(55.39%) gastric carcinoma tissues and in 21 (36.21%) of 58 corresponding para-carcinoma tissues.There was significant difference between the two groups(x~2=5.042,P<0.05).③PCNA LI of 58 gastric carcinoma tissues and the corresponding para-carcinoma tissues were 38.699±10.362 and 22.159±9.532 respectively.The PCNA LI had a significant difference between the two groups(t=8.95, P<0.01 ).PCNA LI of c-myc positive group and c-myc nagetive group was 51.645±9.172 and 32.013±4.916,respectively.There was significant difference between c-myc positive group and c-myc nagetive group(t=9.687,P<0.01).PCNA LI of bcl-2 positive group and bcl-2 nagetive group was 53.007±7.268和29.417±5.865,respectively.There was significant difference between two groups(t=13.602,P<0.01).④AI of 58 gastric carcinoma tissues and the corresponding para-carcinoma tissues was 1.7948±0.6021 and 3.4526±0.5017,There was significant difference between the two groups(t=16.110,P<0.01 ). The number of apoptotic cells was significantly lower in bcl-2-postive(1.5793±0.6946) tumors than bcl-2-negative tumours(2.0103±0.6842,P<0.05).AI of c-myc positive group and c-myc nagetive group was was 1.419±0.549 and 2.290±0.726,it was much lower in c-myc positive group than c-myc nagetive group(t=5.206,P<0.01).Conclusions The abnomal expression of bcl-2,PCNA and c-myc protein is related to the oncogenesis of gastric carcinoma.Bcl-2 protein may play an important role through suppressing cell apoptosis in the pathogenesis of gastric carcinoma.The abnomal expression of c-myc and bcl-2 protein is related to PCNA abnormal accumulation in gastric carcinoma tissue,c-myc and bcl-2 protein could induce proliferation of tumor cells.
