The activity of caspase-3 and caspase-9 was determined by fluorescent analysis. The same methods ware used to observe proliferation,early apoptosis and the activity of caspase-3 and caspase-9 of HaCaT dealed by curcumin and UVB.Results:â‘ After HaCaT were treated with curcumin alone (<2.5umol/L), cell growth,early apoptosis and the activity of caspase-3 and caspase-9 ware not changed,the difference was not statistically significant(P>0.05). After HaCaT were treated with curcumin ( >5umol/L), cell growth was significantly inhibited in a dose- dependent manner, the percentage of the early apoptotic cells were significantly increased and The levels of Caspase-3 and caspase-9 activity were also increased after the treatment(P<0.05).â‘¡After irradiated by UVB with curcumin(=1.25umol/L), HaCaT proliferation was suppressed(P<0.05) and the apoptotic rate of HaCaT cells enhanced from 4.60% to 12.61% When the concentration of curcumin was 10.0umol/L , the apoptotic rate was up to 38.42%. The levels of Caspase-3 and caspase-9 activity were increased by combination of curcumin with UVB.â‘¢Compare with the the effects of curcumin , the apoptotic rate and the levels of Caspase-3 and caspase-9 activity were significantly increased after irradiated by UVB with curcumin of the same concentration.Conclusion: Curcumin (>5umol/L) inhibites the proliferation of HaCaT and induces apoptosis of HaCaT cells through activation of caspase-9 and caspase-3 .Combination of curcumin with UVB irradiation synergistically inhibites the proliferation and induces apoptosis of HaCaT cells. The activation of caspase-9 and caspase-3 was increased notablely .
