ObjectiveTo investigate the effects of different BaP and UVB alone and in combination on the proliferation and the expression of Caspase-14mRNA and Filaggrin mRNA of HaCaT Cells and the correlation between them and damage on HaCaT Cells from BaP and UVB.MethodsPart one:The HaCaT cells were cultured in vitro; Draw cell growth curve; The cultured HaCaT cells were intervened in UVB of10、30、60、90mJ/cm2and BaP of30、50、80、100μ、mol/L respectively, The survival rate was measured by MTT assay; The Caspase-14mRNA and Filaggrin mRNA expression were measured respectively by RT-PCR after24h of different BaP and UVB intervention.Part two:The cultured HaCaT cells were intervened in combining effect of BaP and UVB and the proliferation of HaCaT cells was detected with MTT assay; The Caspase-14mRNA and Filaggrin mRNA expression were measured by RT-PCR after24h of80μmol/L BaP and60mJ/cm2UVB intervention together.Results:1. The survival rate increased gradually in normal HaCaT cells as time extends. At24h after BaP and UVB intervention together, the survival rate of HaCaT cells was lower than that in control group at the same time point (p<0.05).2. The expression of Caspase-14mRNA and Filaggrin mRNA were lower in control group. The Caspase-14mRNA and Filaggrin mRNA expression was significantly increased at24h after intervention of four different concentration of BaP and four different dosage of UVB compared to that in the normal control group at the same time point(p<0.05). Comparative differences between groups were statistically significant (P<0.05).3. The survival rate of HaCaT cells which was intervened in80μmol/L BaP and 60mJ/cm2UVB was lower than only one.4. The Caspase-14mRNA and Filaggrin mRNA expression was significantly increased after24h of80μmol/L BaP and60mJ/cm2UVB intervention (P<0.05). Comparative differences between groups were statistically significant (P<0.01).Results:1. BaP and UVB may inhibit the proliferation activity of HaCaT cells and cause cells damage.2. The Caspase-14mRNA and Filaggrin mRNA expression increased and were influences each other after24h of different BaP and UVB intervention.3. The Caspase-14mRNA and Filaggrin mRNA of HaTaT cells were more significantly increased than only one and have a dose dependent relationship.
