Effects Of Celecoxib On The Expression Of VEGF-C And Bcl-2 MRNA In Prostate Cancer PC-3 Cell Lines | Posted on:2009-03-22 | Degree:Master | Type:Thesis | Country:China | Candidate:N G Hu | Full Text:PDF | GTID:2144360272956077 | Subject:Surgery | Abstract/Summary: | PDF Full Text Request | Objective To investigate the effect of cyclooxygenase-2 selective inhibitor on the expression of VEGF-C and bcl-2 mRNA in human androgen-independent prostate cancer PC-3 cell lines,and study the anticancer mechanism of COX-2 selective inhibitor.To provide the new method and theory basis for the prostate cancer treating clinically, especially the treatment of androgen-independent prostate cancer.Methods The PC-3 cell lines were cultured routinely and fetched the third generation logarithm growth cell,digested by 0.25%trypsin and divided to four groups:Test groups(The cells were treated with celecoxib at different concentrations-10μmol/L,20μmol/L,40μmol/L) and control group(celecoxib was not used).The PC-3 cells were inoculated in 96-well plate,continued culturing 72 hours under identical condition.The cell specimen were collected to followed experiment.The changes of the growth and shape of PC-3 cells cultured in celecoxib of different concentrations were observed under the optical microscope.The growth inhibition curve of celecoxib was obtained by means of MTT.The PC-3 cells Were collected to the followed experiment,and detected the expression of VEGF-C and bcl-2 mRNA in the four groups by RT-PCR method.The quantity of VEGF-C and bcl-2 mRNA was analysed by gel image analysis system.Results The growth of PC-3 cells which were treated by 40βmol/L celecoxib was inhibited obviously.The value of VEGF-C/GAPDH and bcl-2/GAPDH in 10μmol/L celecoxib group were 0.509±0.057,0.444±0.034 respectively.There were no statistical difference of the VEGF-C/GAPDH and bcl-2/GAPDH value between 10μmol/L celecoxib group and control group(P>0.05);VEGF-C/GAPDH value in 20μmol/L and 40μmol/L celecoxib groups were 0.370±0.063 and 0.263±0.062 respectively, bcl-2/GAPDH value in both the groups above were 0.339±0.047 and 0.272±0.042 respectively,both the value above in the two groups decreased significantly compared with control group(P<0.01).Conclusion Celecoxib could inhibit the growth of androgen-independent prostate cancer PC-3 cell lines.The inhibiting effect on PC-3 cell lines is correlated with the concentration of celecoxib.Celecoxib maybe plays its antitumor effect by inhibiting the expression of VEGF-C and bcl-2 mRNA in prostate cancer PC-3 cell lines.This maybe provide new method for the clinical treatment of prostate cancer.
