Background and ObjectiveGastric cancer in our country is a high incidence of malignant tumors, incidence and mortality reside in the top three, but early gastric cancer without obvious symptoms, most of the patients who appears obvious clinical symptoms is already in advanced stage. Surgical resection is the preferred mode of treatment, with the further development of science and technology, new methods of treatment, such as molecular target to treatment, postoperative chemotherapy, comprehensive treatment, including chemotherapy plays an important role.In recent years, It is found that non steroidal anti-inflammatory drugs (NSAIDs) for gastrointestinal malignancies have the function of inhibiting tumor, but due to non steroidal anti-inflammatory drugs that has increased bleeding, ulcers and other gastrointestinal tract complications, therefore,it is generally not as antitumor drug of choice.On this basis, the selective -2 (COX-2) inhibitor is lack of non steroidal anti-inflammatory drugs associated with gastrointestinal side effects, and has the role of inhibiting tumor.Celecoxib is an epoxy selective cyclooxygenase-2 (COX-2) inhibitor class of classical one of the drugs. the present study shows that, celecoxib promotes apoptosis of tumor cells, inhibits the proliferation of tumor cells.At present,the study focused on colon cancer cells, however, the study of gastric cancer, the relative partial less, its role in cancer cells and the mechanism is still unknown. Therefore, this study aims to investigate:1, celecoxib and 5-FU, can inhibit the proliferation of tumor cells, promote apoptosis;or not.2, Combined use of celecoxib and 5-FU, Effects of VEGF and Survivin expression on tumor growth related genes.Methods:First on human gastric carcinoma BGC-823 cells were cultured in vitro and by MTT method, detection of different concentrations under the effect of celecoxib on different time, effect on the proliferation of BGC-823 cells and calculation of the corresponding IC50 values;By general PCR method for detection of celecoxib,5-FU, and celecoxib combined with 5-FU on BGC-823 cells, VEGF and survivin gene expression and to observe the effects of monotherapy and combination therapy effect.The three groups and the above measurement data using analysis of variance, two sample mean by t test,data to Mean+SD, data processing using SPSS 19 statistical software. To P< 0.05 for a statistically significant difference.Results:MTT results showed that celecoxib and 5-FU have a role in inhibiting tumor, with the extension of time, for enhanced the inhibition of tumor, both are drug synergistic sensitization.l, the IC50 of celecoxib, the same intervention concentrations of celecoxib inhibited the proliferation of BGC-823, the 24 h,48 h,72 h IC50 respectively:(255.36+21.1) umoL/L, (149.12+2.29) umoL/L, (89.95+1.61) umoL/L.2, inhibition rate of each group:the 5-FU group 24h,48h,72h inhibitory rate were (52.13+0.5,67.42+0.3,89.26+0.3); celecoxib group at 24h, 48h,72h inhibitory rate were (24.53+0.2,51.87+0.1,71.37+0.2); celecoxib combined with 5-FU group 24h,48h,72h inhibitory rate (86.89+0.1,91.39+0.2,95.39+0.3), respectively.Ordinary PCR results showed that celecoxib and 5-FU has inhibited the expression of VEGF and survivin gene, in the effect of combined application had the strongest inhibitory effect, between the statistical results show obvious differences.Conclusion:Celecoxib inhibits the proliferation of human gastric cancer cell line BGC-823. With the concentration increasing and time prolonging, for enhanced inhibition of tumor cells;Combined use of celecoxib and 5-FU, to strengthen the inhibition of tumor cells significantly.Celecoxib can inhibit the expression of VEGF and Survivin in human gastric cancer cell line BGC-823, Combined with 5-FU can synergistically inhibit the proliferation of gastric cancer cells.
