The Study On The Correleation Between Urokinase-type Plasminogen Activator And Plasminogen Activator Inhibitor-1 And The Invasion And Metastasis Of Endometrial Carcinoma | Posted on:2009-06-27 | Degree:Master | Type:Thesis | Country:China | Candidate:L Meng | Full Text:PDF | GTID:2144360272956349 | Subject:Obstetrics and gynecology | Abstract/Summary: | PDF Full Text Request | Objective:To characterize the differential expression of urokinase-type plasminogen activator(uPA) and plasminogen activator inhibitor-1(PAI-1) mRNA in endometrial carcinoma,and their significance.To investigate the variation of the plasma concentration ofurokinase -type plasminogen activator(uPA) and plasminogen activator inhibitor-1(PAI-1) in endometrial carcinoma.Methods:The expression of uPA and PAI-1 gene in 35 cases of endometrial carcinoma and 18 cases of endometrial hyperplasia and 16 cases of normal endometrial tissues was examined by semi-quantitative RT-PCR.The levels of uPA and PAI-1 in 35 patients with endometrial carcinoma and 18 endometrial hyperplasia and 16 normals were measured by ELISA.and the relevant clinical pathological parameters were analyzed.Results:(1) The relative contents of uPA mRNA and PAI-1 mRNA and uPA/PAI-1 in endometrial carcinoma tissue were significantly higher than in endometrial hyperplasia tissues and normal endometrial tissues(P<0.01).The expression of uPA and PAI-1 gene in endometrial hyperplasia patients were not significantly higher than in normal group(P>0.05).The expression increased with the increase of surgical-pathological stage clinical stage and histological grade(P<0.05,respectively). With the deeper myometrial invasion of endometrial carcinoma,the more uPAmRNA and PAI-1mRNA was expressed(P<0.05).More uPAmRNA and PAI-1mRNA was expressed in patients with lymph metastasis than not(P<0.01).The relative contents of uPA and PAI-1 mRNA in patients with endometrial carcinoma did not correlate with pathologic typing(P>0.05).(2) The concentration of uPA and PAI-1 and uPA/PAI-1 in the plasma of endometrial carcinoma patients were significantly higher than in the plasma of endometrial hyperplasia and normal endometrial patients(P<0.01, respectively).In endometrial hyperplasia patients the levels of plasma uPA and PAI-1 were not significantly higher then in normal group(P>0.05).The relative contents increased with the increase of surgical-pathological stage clinical stage and histological grade(P<0.05,respectively).With the deeper myometrial invasion of endometrial carcinoma,the more uPA and PAI-1 was meaesured(P<0.01).More uPA and PAI-1 was eaesured in the plasma of patients with lymph metastasis than not(P<0.01).The relative contents of uPA and PAI-1 in the plasma of patients with endometrial carcinoma did not correlate with pathologic typing(P>0.05).
