ObjectiveTo investigate the expression and significance of MMP-9 and TIMP-1 on the formation of postoperative intestinal adhesions.MethodsRat models of intestinal adhesions were established with ileal abrasion, absolute alcohol dropped in wound surface and arteria mesenterica of cecum clamped. Formation of postoperative intestinal adhesions was observed by naked eye and light microscope. The expression of MMP-9 and TIMP-1 protein in the control ileum and adhesive tissues was determined by immunohistochemistry assay and image analysis.Results1. Visual inspection:Compared with the 2th day after injured, intestinal adhesions in the 7th and 14th day after injured were accrescenced(P<0.05).While intestinal adhesions in the 7th and 14th day after injured had no significantly difference(P>0.05).2. Light microscope observation:A consecutive coat of mesothelial cells was observed in the serous membrane of control group. There was no consecutive coat of mesothelial cells in the serous membrane,but apparentely inflammatory reaction occurred in the 2th day after injured. Loosened collagen fibrils and profuse fibroblasts were observed in the serous membrane in the 7th day after injured. In the 14th day after injured,there were less fibroblasts,more compact collagen fibrils and a consecutive coat of mesothelial cells.3. Compared with the control group , the expression of MMP-9 protein in the 2th day after injured increased significantly(P<0.05). Compared with the control group and the 2th day after injured, the expression of MMP-9 protein in the 7th and 14th day after injured decreased significantly(P <0.05). 4. The expression of TIMP-1 protein in the 2th ,7th and 14th day after injured was all higher than that in the control group(P<0.05).While the expression of TIMP-1 protein in the 2th ,7th and 14th day after injured had no difference(P>0.05).Conclusions1. Rat models of intestinal adhesions established with ileal abrasion can be used to study the formation of postoperative adhesions.2. MMP-9 can promote the formation of intestinal adhesions in the early phase of postoperation.3. TIMP-1 is closely related to the formation of postoperative intestinal adhesions in the late phase.
