Endometrial carcinoma is one of the common malignant tumors in female.The incidence is climbing year by year in recent years.TypeⅠendometrial carcinoma is estrogen-depended,consisting of 80%-90%of endometrial carcinoma.There are articles reporting that typeⅠendometrial carcinoma can be induced by estrogen persistently acting on endometrial cells.As the development of research on stem cell,scientists have found that certain features of carcinoma can be explained with the theory of stem cell.Reports showed that a limited number of cells which have similar features with stem cells exist in carcinoma tissue and have a relationship with the growth and metastasis of carcinoma.In the process of inducing endometrial carcinoma by estrogen,stem cells may contribute to it in the following two aspects:on one hand,it may involve in the genesis of carcinoma,on the other hand,it may prevent endometrial cells from cancerization because of immune reconstitution.In order to investigate the role of bone marrow stem cells in the process of inducing endometrial carcinoma by estrogen,I made this elementary study.Methods:1.Bone marrow cells suspension from male,transgenic,green fluorescent protein (GFP) mice were obtained.2.The bone marrow cell suspension/DMEM was injected into wild-type(wt) female mice after Co60 systematic radiation.Subsequently,the survival mice were fed with or without 17β-estradiol(E2) supplemented water for 4 months.3.After 4 months,the recipient mice were sacrificed to obtain the blood,uterine and bone marrow.4.The blood serum estrogen was detected.The specimens were detected with FCM for the expression of GFP in bone marrow cells,fluorescent light microscope for GFP's green fluorescent light observation and HE staining for histopathologic evaluation.Results:1.Blood serum estradiol assessment showed that the difference of estradiol level in groups with estrogen supplement is not significant,and the estradiol level is higher than the normal group.2.Consequently,94.78%+/-2.5%of bone marrow cells of model with bone marrow transplantation after systematic radiation expressed GFP by fluorescence and FCM compared with those of control,which indicated success of BMT.3.Macroscopic appearance and histopathology show that model with bone marrow transplantation after systematic radiation cannot be induced carcinoma,while model with only E2 supplement can be induced carcinoma.Conclusion:In my study,we haven't got the evidence for inducing bone marrow stem cells to differentiate into endometrial carcinoma cells by supplying with estrogen. Contrarily,we found that bone marrow stem cells transplantation of allochthonous after destroying original bone marrow stem cells can be an inhibitor of inducing endometrial carcinoma by supplying with estrogen.
