PARTⅠ:The effect of electro-acupuncture and physical training on the infarct volume and neurological behavior after focal brain ischemia in rats.To investigate whether electro-acupuncture and physical training could improve neurological function and reduce the infarct volume after focal brain ischemia.Method:Thirty-two male adult Sprague-Dawley rats(2~3month old,n=32) were subjected to 60 min right middle cerebral artery occlusion(MCAO).All rats were randomly assigned to one of the four groups:electro-acupuncture group, physical training group,control group and sham operation group.There is 8 rats in each group.Electro-acupuncture group underwent 30 rain electro-acupuncture treatment while physical training group underwent 30 min treadmill training per day for 2 weeks 24h after MCAO procedure.The neurological score and infarct volume were evaluated 2 weeks after MCAO.Result:After two weeks,the neurological score of electro-acupuncture group and physical training group are heigher when compared with spontaneous recovery (P<0.05).The infarct volume in electro-acupuncture group and physical training group are smaller when compared with that in controls(P<0.05).But there is no significant difference between electro-acupuncture group and physical training group.Conclusion:Electro-acupuncture and physical training can improve neurological function and reduce the infarct volume afert focal brain ischemia.PARTⅡ:The effect of electro-acupuncture and physical training on the expression of Angiopoietin-1 and its receptor Tie-2 after focal brain ischemia in rats. To investigate whether electro-acupuncture and physical training can up-regulate the expression of Angiopoietin/Tie pathway after focal brain ischemia.Method:Fifty-six male adult Sprague-Dawley rats(2~3month old,n=56) were subjected to 60 rain right middle cerebral artery occlusion(MCAO).All rats were randomly assigned to one of the four groups:electro-acupuncture group, physical training group,control group and sham operation group.There is 14 rats in each group.24h after MCAO procedure,electro-acupuncture group underwent 30 min electro-acupuncture treatment while physical training group underwent 30 min treadmill training per day for 2 weeks.Western bloting was used to evaluate the expression of Angl/Tie2.Result:After two weeks,the neurological score of electro-acupuncture group and physical training group are heigher when compared with spontaneous recovery (P<0.05).The expression of Angiopoietin-1 and its receptor Tie-2 were up-regulated in the electro-acupuncture group and physical training group(P<0.05)when compared with controls.But there is no significant difference between the electro-acupuncture group and physical training group.Conclusion:Electro-acupuncture and physical training can up-regulate the expressin of Angiopoietin/Tie system after focal brain ischemia,leading to the inproving of neurological function.PARTⅢ:The effect of electro-acupuncture and physical training on the PI3K/Akt signal transduction pathway after focal brain ischemia in rats.Object:To investigate whether electro-acupuncture and physical training can activate the PI3K/Akt signal transduction pathway after focal brain ischemia,leading to the reduction of endothelial cell apoptosis.Method:Thirty-two male adult Sprague-Dawley rats(2~3month old,n=32) were subjected to 60 rain right middle cerebral artery occlusion(MCAO).All rats were randomly assigned to one of the four groups:electro-acupuncture group, physical training group,control group and sham operation group.24h after MCAO, electro-acupuncture group underwent 30 min electro-acupuncture treatment while physical training group underwent 30 min treadmill training per day for 2 weeks. Western bloting was used to evaluate the activation of PI3K/Akt pathway and cell apoptosis 2 weeks after MCAO.Result:After two weeks,the neurological score of electro-acupuncture group and physical training group are heigher when compared with spontaneous recovery (P<0.05).The phosphorylation level of PI3K/Akt in the physical training group is higher when compared with control groug(P<0.05);while the expression level of Bax in physical training group is lower when compared with control group(P<0.05). But there is no big difference in electro-acupuncture group when compared with controls.Conclusion:Physical training can activate PI3K/Akt signal transduction pathway,which may be associate with the neurological recovery.But electro-acupuncture may improve neurological recovery through another way.
