ObjectiveOn the basis of comprehensive therapy with acupuncture for apoplexyat the present time, combining modern pharmacological research to preparecollagen tetramethylpyrazine sustained-elease dose, to research theeffect of embedding medicine in Dazhui and Neiguan acupoints on the infarctvolume and its regulating effect on matrix metalloproteinase-9 and itsinhibitors-1 after focal cerebral ischemia-reperfusion.MethodsTotally 160 healthy SD rats were selected and divided randomly into6 groups, The groups included normal group, sham group, model-24h group,model 120h group, model 120h group, abdominal cavity injection 24h group,abdominal cavity injection 72h group, abdominal cavity injection 120h group,electroacupuncture 24h group, electroacupuncture 72h group,electroacupuncture 120h group, embedding medicine in acupoints 24h group,embedding medicine in acupoints 72h group and embedding medicine inacupoints 120h group with 10 in each group. The model of ischemiareperfusion of cerebral middle artery was established by the modifiedthread occlusion method, then embedding medicine in Dazhui and Neiguanacupoints with collagen tetramethylpyrazine sustained-release dose. 4 ratsof each group were decollated directly to extract unsalted brain tissuewith TTC chromating; and other 6 rats were reperfused with 4%paraformaldehyde after being anaesthetized by 10%chloral hydrate, and thentaken out the brain rapidly. The brain tissue 2mm away from the chiasmawas cut frontally, the middle part left, immersed in hard paraffin. Usedimmunohistochemistry SABC method to investigate the expression of MMP-9and TIMP-1, and the effect of all the above index of electro-acupuncturewere observed. The entire datum was analyzed by statistics.Results1. MCAO and reperfusion injury can damage the rats'activity distinctly.The nerve symptoms of abdominal cavity injection group, electro-acupuncture group and embedding medicine in acupoints group wereameliorated obviously; there was a significant difference between modelgroup and each therapy group, and the embinding medicine in acupoints groupcan improve rats' nerve sympotom remarkably furthermore(compared withabdominal cavity injection group, P<0.01; compared withelectro-acupuncture group, P<0.05).2. There was visible infarct focus in ischemical area after focalcerebral ischemia-reperfusion by TTC chromating. Compared with model group,the methods of abdominal cavity injection, electro-acupuncture andembedding medicine in acupoints can lighten cerebral infarct volume, andthe method of embedding medicine in acupoints was more ascendant (comparedwith abdominal cavity injection group, P<0.01; compared withelectro-acupuncture group, P<0.05).3. The expression of MMP-9 heightened and the expression of TIMP-1depressed in brain tissue after focal cerebral ischemia-reperfusion withthe method of immunohistochemistry. Compared with model group, theabdominal cavity injection and electro-acupuncture can down-regulate MMP-9'expression and up-regulate TIMP-1' expression, but the regulation effectof MMP-9 and TIMP-1 was not clear with embedding medicine in acupoints 24hgroup (compared with model group, P>0.05; compared with abdominal cavityinjection group, P>0.05). The preponderant effect of embedding medicinein acupoints group represented at 72h and 120h (compared with abdominalcavity injection group and electro-acupuncture group, P<0.05).ConclusionThe expression of MMP-9 increased and the expression of TIMP-1 reducedin ischemical brain tissue after focal cerebral ischemia reperfusion inrats, which destroyed the structure' integrality of cerebral microvas andincreased brain infarct volume, and then aggravated ischemia reperfusioninjury.
