Objective: The leaves of B. papyrifera (Linn.) Vent are also called Chu leaf, Guhuang leaf and Jianghuang leaf ect. Tasting sweet and innocuity, having the function of removing pathogenic heat blood and inducing diuresis, so it can treat hematemesis, blood collapse, hemorrhage for traumatism, edema, hernia, diarrhea, tinea and sore ect. It distributes many places in China, the resources are very rich. The decoction of the leaves of B. papyrifera (Linn.) Vent is used in local folk medicine against chronic prostatitis in Henan Province of China, which function had been proved by pharmacology experiment. The phytochemical studies of the plant have few been reported previously. In order to search the bioactive constituents, we carried out a systematically phytochemical study on the leaves of B. papyrifera (Linn.) Vent. Methods: The dried leaves of B. papyrifera (Linn.) Vent were extracted with 50% aqueous acetone twice at room temperature. The extract was concentrated under reduced pressure at 50℃to afford a dark-brown residue, which was suspended in water, and extracted with ether, ethyl acetate, n-butanol, respectively. The eher and ethyl acetate layers were chromatographied on a silica gel column using a gradient system of petroleum ether:EtOAc and CHCl3:MeOH as eluent. The n-butanol layer was chromatographied over macroporous absorption resin Diaion HP-20 respectively with aqueous methanol gradiently. The eluates from resin were repeatedly chromatographied on Sephadex LH-20, Toyopearl HW-40C, Silica gel columns. Results: 35 compounds have been isolated from the leaves of B. papyrifera (Linn.) Vent by various chromatographic methods. The structures of 29 compounds have been elucidated by spectroscopic and chemical methods, as heptadecanoic acid (LHW-3),β-sitosterol (LHW-5), apigenin ( LHW-8 ), luteolin (LHW-9), ficustriol (LHW-10), 3β-hydroxy-5α, 6α-epoxy-β-ionone-2α-O-β-D-glucopyranoside(ELHW-8), (6S,9S)-6- hydro xyl-3-oxo-α-ionol-9-O-β-D-glucopyranoside(ELHW-6), (2R,3R,5R,6S,9R)-3 -hydroxy-5,6-epoxy-β-ionol-2-O-β-D-glucopyranoside(ELHW-4), (2R,3R,5R,6S,9R) -3-hydroxy-5,6-epoxy-acetyl-β-ionol-2-O-β-D-glucopyranoside(ELHW-3), apigenin- 7-O-β-D-glucopyranoside (ELHW-14), chrysoerid-7-O-β-D-glucopyranoside (ELHW- 12), apigenin-7-O-β-D-glucopyranuronide(ZLHW-2), vitexin-7-O-β-D-glucopyranosi -de(ZLHW-8), isoorientin(ZLHW-10), 5,7,4'–trihydroxyl-6-C-[a-L- Rhamnopyranosyl -(1→2)]-β-D-glucopyranosyl flavone(ZLHW-9), 5,7,4'-trihydroxyl-8-C-[a-L-Rhamno -pyranosyl (1→2)]-β-D-glucopyranosyl flavone (ZLHW-9'), 2′-deoxyadenosine (ELH W-13), thymidine(ELHW-2), 2′-deoxyuridine(ELHW-7), lariciresinol-9-O-β-D- glucopyranoside(ELHW-11), 4-hydroxycinnamicacid-9-O-β-D-gluco-pyranoside(EL HW-9), (-)-syringaresinol-4-O-β-D-glucopyranoside (ELH W-1), p-hydroxyace -tophenone (ZLHW-6), icariside B1 (ELHW-15), 3- hydroxy-5α,6α-epoxy-β-iononoe (LHW-7), benzyl benzoate-2,6-di-O-β-D-glucopyranoside (ZLHW-1), vitexin (ELHW -17), 1,2,3-trihydroxybenzene (ZLHW-12), broussolactone (ZLHW-4). Conclusion: Among them, there are 3 new compounds, including ZLHW-1, ZLHW-2 and ELHW-3. All the compounds are obtained from B. papyrifera (Linn.) Vent for the first time.
