Objective: Our country is one of the most abundant plant resources countries, but come to the trade in the markets, it is share a smaller market portion. Therefore, it is need to vigorously promote the development of medicine resources in modern research. Including apply to the modern technology to research, extract of active ingredients, evaluate effectiveness, and to develop of natural preservatives and cosmetics, thus it has a better use of plant resources. The objective of this study is to extract the natural active component from Broussonetia Papyrifera leaves, and determine the active component with HPLC to approach the effect of action of anti- Epidermophyton floccose and natural preservative. In order to get rational development and utilization of Broussonetia Papyrifera leaves in our country.Methods: The Broussonetia Papyrifera leaves picked in Huanggang district of Hubei Province were dried, then extracted by 75% alcohol for 24h, repeated 3 times, collected mixed liquor.(1) The mixed liquor is extracted by DAB–6,hydrolysised filtered, et al. then determine the flavanoid with HPLC.(2) The mixed liquor is saponified by alcohol and base, extracted by DAB-6 (refining), concentrated decompress, chromatographic fractionation, filtration, et al. and then determine coenzyme Q10 with HPLC.(3) Drug culture medium flat plate method is carried out anti-mycology test of Thichophyton rubrum, Thichophyton mbrum, Epidermophyton floccosum and Candidadlbicans using the extracted flavonoid.(4) Add the extracted active component to soy sauce, and set stand control to observe preservation effect in qualitation and quantity, moreover carry out a adding-bacterium for 28 days microbe Challenge test. Results: (1) according to HPLC, the flavonoids in Broussonetia Papyrifera leaves including meletin,isoliquiritigenin and daucosterol,whose contents are 6.51g,7.06g,1.74g. The whole content of flavonoids is 15.31g. the contents of meletin, isoliqui- ritigenin and daucosterol possess fine linear correlation during the circumscription of 3.1~25.0μg/mL,2.5~50.0μg/mL,2.5~50.0μg/mL.The average spotting recovery rate (e.g: meletin) is 99.91%. RSD is 0.91%(n=6).(2) The average content of coenzyme Q10 extracted from 1.0kg Broussonetia Papyrifera leaves is 0.66g, which possess fine linear correlation during the circum scription of 4.0~64.0μg/ml(r=0.9997). The average spotting recovery rate is 98.66%, RSD is 0.97%(n=6). the LDL of CoQ10 in Broussonetia Papyrifera leaves is 10ng/ml (S/N =3).(3) The anti-mycological tests of Thichophyton rubrum, Thichophyton mbrum, Epidermophyton floccosum and Candidadlbicans indicate that Broussonetia Papyrifera leaves possess good anti-mycetes effect, the anti-superficial mycetes test of extracts according to different solution indicate they have good anti-mycetes effect. The minimal inhibitory concentrations of the extracts are lower than MIC of received anti-mycological drug now.(4) The flavonoids of Broussonetia Papyrifera leaves have a good preservative effect. Cultivate in the 37℃and room temperature for 14 days, we find inhibition of bacterium and mould under concentration of 0.5% and higher. In the microbe Challenge test, 1.0% extract of Broussonetia Papyrifera leaves can get through microbe Challenge test, whose effect is better than 0.1% sodium benzoate. According to the test, the extract of Broussonetia Papyrifera leaves have a good bacteriostasis and preservative effect.Conclusion: To determine the flavonoids and coenzyme Q10, HPLC is sensitive, fast, accurate, it can be used to analysis the flavonoids and coenzyme Q10 in extract of Broussonetia Papyrifeia leaves. The rudiment bacteriostasis and preservative test indicates the active component of Broussonetia Papyrifera leaves—flavonoids possess determinate bacteriostasis and preservative effect. it makes the foundation for exploitation of plant of medical resource in our country. The resource of Broussonetia Papyrifera leaves is abundant, they can be used as broad-spectrum anti-superficial mycetes Chinese herbal medicine. It is expected to develop a kind of new anti-mycetes Chinese herbal medicine. And as a kind of natural antiseptics, the flavonoids of Broussonetia Papyrifeia leaves is worth of exploitation.
