Objective:To dynamically supervise the changes of the central cholineragic system of parkinson's disease rats in various stages of the pathogenosis, to find out the rules of the changes,to try to investigate its mechanism, and to make a model of treatment for the further reseach,to try to investigate its mechanism.Method:Striatum lesioned rat models of Parkinson disease with 6-OHDA were treated with levodopa for 7 days afer the successful models were made.Per seven days during the course, the Ache-positive fiber in Meynert nuclei were detected by Ache histochemical and choline acetyl transferase (ChAT)-positive neurons were detected by immunohistochemistry.HPLC was used to measure the concentration of Ach in the striatum in live PD rats. The the M2–acetylcholine receptor andα4-acetylcholine receptor were detected by immunohistochemistry.Results:The Ache-positive fiber density and the number of ChAT–positive cell in Meynert nuclein decreamed in the course. The concentration of Ach in striatum decreased progressively in the course. Meanwhile the M2–acetylcholine receptor increased andα4-acetylcholine receptor collapsed . After the treatment with levodopa, none of all was changed obviously.Conclusion : The central cholinergic cells decreamed in PD rats. The concentration of Ache decreased. The M2-R increased, meanwhile theα4-R collapsed. The dysfunction of central cholinergic system cannot be reversed by levodopa.
