| Objective:To investigate its protective effect and action mechanism of Tetramethylpyrazine (TMP) injection after treatment in rats with acute spinal cord injury.Methods: Selected 84 adult Sprague-Dawley rats were randomly assigned into 3 groups: normal control group(n=2×6),spinal cord injury group(n=6×6) and TMP treatment group (n=6×6). The rats in spinal cord injury group and TMP treatment group rats were enrolled to establish moderate spinal cord injury models with Allen's method, in which they received physiological saline and Tetramethylpyrazine injection treatment respectively.The rats were sacrificed at 8h,1d,3d,1w,2w,3w after surgery, then behaviour score and tiltboard test were conducted, the tissue sections were stained with hematoxylin and eosin,immunohistochemistry and terminal deoxynucleotjdyl transferase-mediated dUTP nick end labeling method.Results: All rats were involved in the result analysis. There were significant differences in behaviour scores between TMP group and Spinal Cord Injury group at 1w,2w,3w (P<0.05). There were significant differences in tiltboard largest angle between TMP group and Spinal Cord Injury group at 1d,3d,1w,2w,3w (P<0.05). The histopathology changes in spinal cord tissue observed with HE staining microscopic examination in TMP group was better than that in injury control group(P<0.05).Expression of iNOS and caspase-3, apoptotic cells were observed in the three groups,and there were significant differences in the expression of iNOS and caspase-3, and apoptotic index between Spinal Cord Injury group and TMP group at 8h,1d,3d,1w,2w,3w (P<0.05).Conclusions: TMP injection can inhibit expression of iNOS,caspase-3 and apoptosis after spinal cord injury in rats,TMP injection has a protective effect on the recovery of the nerve function.
