Association Between GPRA Gene Polymorphisms And Susceptibility Of Allergic Asthma

Objective: To investigate whether SNP563704, SNP522363 and SNP546333 of the GPRA(G Protein related receptor for asthma) gene are associated with asthma in Han nationality of Hubei province in China, and the association of the haplotypes with asthma.Methods: The polymorphisms are detected with polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP), and total IgE levels with chemiluminescent analyzer.Results: (1) The genotype frequencies of SNP563704 were 0.324 for C/C,0.524 for C/T, 0.152 for T/T in allergic asthma patients in a Chinese population. The genotype frequencies of SNP522363 were 0.289 for C/C, 0.521for C/G, 0.190 for G/G in allergic asthma patients in a Chinese population. (2) No significant differences in the distributions of SNP563704 and SNP522363 polymorphisms were found between allergic asthma and healthy control subjects(χ2=1.880,P>0.05;χ2=0.700,P>0.05, respectively). (3) No significant differences in genotype of SNP563704 and SNP522363 and serum total IgE were found in allergic asthma patients(F=0.714, P>0.05;F=0.083, P>0.05, respectively). (4) The distribution of frequencies of 4 haplotypes showed significant difference(χ2 = 16.50, P<0.01). The haplotype frequencies of CT and GC were higher in asthma subjects than that of in controls. (5) Compared with German population , there are significant differences in the genotypes and alleles distributions of the SNP563704 (χ2=14.260,P=0.001;χ2=11.757,P=0.001),but compared with Anhui population, it is similar in the distributions of the SNP(χ2=1.427,P=0.490;χ2=0.074,P=0.786). (6) We haven't detected the polymorphism of SNP546333.Conclusion: The polymorphisms of SNP563704 are not associated with serum total IgE and asthma in Han nationality in Hubei Chinese population. However, the genotypes and alleles distributions of SNP563704 are significantly different among ethnics. The polymorphisms of SNP563704 and SNP522363 are not associated with allergic asthma in Han nationality in Hubei Chinese population. However, the haplotypes are associated with allergic asthma.
