| Objective:Single-nucleotide polymorphism (SNP)-based genome-wideassociation study revealed that markers in or around ORM1-like3(ORMDL3) gene were related to childhood asthma, And the relevance wasconfirmed in distinct ethnical groups. But the replication studies to definethe genetic associations between markers in or around ORMDL3gene andchildhood asthma in Chinese population are inadequate. Our study wasdesigned to investigate the genetic associations of single-nucleotidepolymorphisms in and around ORMDL3gene with childhood asthma inChinese population.Methods:Peripheral venous blood (2mL) was drawn from352persons (152patients (5.872±2.543years) with childhood asthma and190healthycontrols (6.038±2.526years)), and the genomic DNA was extracted fromthose blood samples. Primers for PCR and single base extension were designed by using Assay Designer software package (Sequenom). SNPgenotyping was performed with MassARRAY system (Sequenom) bymeans of matrix assisted laser desorption ionisation-time of flight massspectrometry (MALDI-TOF-MS) method.Results:Among all six SNPs, only the genotype frequencies of SNP rs7216389were found to be significantly different between asthmatic children andcontrols (P=0.018), and asthmatic children presented with a higher portionof T allele frequencies (P=0.04, OR=1.653,95%CI=1.170-2.333) in SNPrs7216389, compared with controls. The TT genotype of SNP rs7216389was found as a risk factor for childhood asthma by logistic regressionanalysis (P=0.016, OR=1.704,95%CI=1.105-2.628). There was nosignificant association between the TT genotype of SNP rs7216389andclinical features of childhood asthma.Conclusion:Taken together, results from our current study highlight the role ofORMDL3as a susceptible gene for childhood asthma, and additionally, theTT genotype of SNP rs7216389polymorphism may increase thesusceptibility to childhood asthma among Chinese population. |
