Effect Of Leptin On The Expression Of Cyclin D1,CDK2 Of Vascular Smooth Muscle Cells

Object: To explore the effect of leptin on cyclinD1,CDK2 and cell cycle phases of vascular smooth muscle cells cultured in vitro and study the possibility application of leptin on the atherosclerosis.Methods: The effect of leptin on VSMC proliferation was studied by using MTT colorimeter, the cell cycles of the groups were analyzed by using flow cytometer,cyclin D1,CDK2 expressions were examined by the method of Western-blot.Results: Compared with the control group ,the percentage of the cells of the leptin group in G0/ G1 phase decreased ,while that in S and G2/M phase increased ; the amount of protein expression of cyclin D1,CDK2 was increased depending on the dose of the leptin(P<0.05). Compared between the 100ng/ml and the 200ng/ml the results had no statistics difference (p>0.05).Conclusion:Leptin can promote PI index of the VSMC and the expression of cyclin D1,CDK2 protein , promote the proliferation of VSMC.
