Objective :To prepare the primary culture of rat vascular smooth muscle cell and vascular endothelia cell and compare their cell cycle and biological characteristics.Methods :Endothelial cells and smooth muscle cells were isolated from Sprague-Dawley rat arteries and cultured in medium containing FBS. Next, the cell cycle properties of the cultured cells were evaluated by FACS; The expression of cyclinD1,cyclinE,CDK2 cyclinD1,cyclinE,CDK2,P27and PCNA were also investigated by Western Blot.Results :The normal cultured aortic endothelial cells and smooth muscle cells has no significant difference in cell cycle distribution as well as the level of cyclin D1,cyclinE,CDK2,P27 and PCNA,P27 and PCNA. Paclitaxel and Roscovitine can attenuate the proliferation of both of them without any selectivity.Conclusion:There are no differences of the cell cycle distribution between rat aortic endothelial cells and smooth muscle cells.
