Objective: TO investigate the effect of extract of ginkgo biloba leaf (EGB)on the apoptosis of retinal ganglial cell in diabetic rats , and discuss the mechanism of EGB preventing and curing diabetic retinopathy . so provide the theoretical basis for its clinical uses.Methods: 30 healthy adult Wistar rats were chosen and divided into three groups of control group randomly , model group, and EGB therapia group. Diabetic rats were induced by intraperitoneal injection of streptozotocin ( STZ). EGB therapia group was lavaged with EGB solution for 12 weeks . TUNEL method, SP method and computer– picture analytical system were used to observe the apoptosis, Electron microscope was used to observe the ultrastructure change of ganglia cells.Results: The apoptosis index of ganglial cells in model group increased obviously compared with the control group, while it decreased in the EGB therapia group compared with the model group and the distinction had statistical significant (P<0.01). The expression of Bax and Bcl-2 in ganglial cells was upregulated in the model group compared with the normal contrast group, however, the expression of Bax was downregulated in the model group compared with the EGB therapia group. The expression of Bcl-2 was upregulated in the model group compared with the EGB therapia group. The distinction also had statistical significant (P<0.01). Damage of the ultrastructure of retinal ganglial cells in the EGB therapia group was lightened obviously.Conclusions: EGB can inhibit the apoptosis of retinal ganglial cells in diabetic rats and can lighten the damage on the ultrastructure of retinal ganglial cells obviously. We presume that EGB could influencethe expression of apoptosis -related genes Bax and Bcl-2 in retinal ganglial cells. This may be its anti-apoptosis mechanism.
