Diabetes mellitus is one of the common metabolism disease which can seriously destroy human's health. It is our advantage to prevent and treat diabetes mellitus with TCM. Huanglianxiaoke Granule is based on the Xiaoke pill of beijiqianjinyaofang authored by the medical sage Sun simiao, the Tang Dynasty. It has a less medical ingredients but its effect is special and conspicuous. Huanglianxiaoke Granule has a main therapeutic method to strengthen qi, nourishing yin, which is according to the pathogenic characteristic, however, the research about Huanglianxiaoke Granule is not enough. The main contents of this issue are as follows.(1) The extraction technology for Huanglianxiaoke Granule was investigated through single comparative and orthogonal law with the use of the extraction rate of berberine hydrochloride and polysaccharide as the assessment index. Results: After soaked for 30min, the coarse powder of Copis chinensis Franch was decoted with 12 times water for 1.5h one time; the four kinds of raw materials were decocted together with the residue of Copis chinensis Franch, and after soaked for 20min, for 1.5h just one time with 12 times water.(2) The mice were injected with epinephrine ( 250mg/kg·BW ) for hyperglycemic model, The blood glucose levels in various animal models were determined with the models of hyperglycemic mice induced by epinephrine and glucose and also normal mice, and the effects on blood glucose levels were observed, then the toxicity of Huanglianxiaoke Granule was examined. Results: Huanglianxiaoke Granule can decrease the blood glucose levels of hyperglycemic mice induced by epinephrine, but had no significant effect on normal mice.(3) The mice were injected with alloxan for diabetes model, to investigate the he effects of the dose, the way and times of adminstraion of alloxan on the model of diabetes induced by alloxan in mice. The experimental results showed that the mortality would be increased as the dose of alloxan and the body weight of mice were increased. The mortality rate was reduced when alloxan was given in several times at the same dose. In addition, the incidence of diabetes by intravenous administration of 60mg/kg·BW alloxan was higher than that by intraperitoneal injection of 200mg/kg·BW alloxan. And then investigating the effect of Huanglianxiaoke Granule on diabetic mice induced by alloxan. The level of weight and the amount of watering and eating were recorded everyday and the level of fasting blood glucose, carbohydrate tolerance, urine glucose, Serous biochemical indicator, liver glycogen, index of thymus, index of spleen were detected at the end of treatment. Results: compared with the model group, Huanglianxiaoke Granule can improve the symptom of diabetes mellitus in mice, decrease the fasting blood glucose, improve the carbohydrate tolerance abnormality of diabetic mice, increase the liver glycogen, enhance the index of thymus and spleen, which have the same function with the positive drug.(4) experiment of acute toxic mechanism :the mice were respectively gave the highest dosage which were 50g medicinal /kg weigh of Huanglianxiaoke Granule in one day. The maximum tolerance dose of Huanglianxiaoke Granule is 100 higher than clinical recommending doses which proves to be safe.
