Study On Immunity Pharmacodynamics And Correlation Mechanism Of Recombinant Human Thymosin Alpha-1 By Injection

Object:The purpose of this experiment is to detect the effects of immunologic suppression mouse and normal mouse's pharmacodynamics and immunogenicity which caused by thymosin alpha-1(Tα1) in injection。Method:We used the methods of macrophage swallowing neutral red, 3-(4, 5-dimethylthiazol-2yl)-2, 5-diphenyl tetra- zolium bromide (MTT) and ELISA assay to detect the ability of mouse's macrophage phagocytosis, the ability of T, B lymphole-ukocyte transformation, cytotoxic effector potential caused by natural killer cell(NK) and the level of IL-2. In addition, we also measured the body weight, the spleen and thymus weight of mice and calculated the spleen, thymus index. In the experiment to detect the effect of rh-Tα1 to the immumosuppression mouse, cyclophosphamide was used to make the model of immumosuppression mouse.Result:In the immumosuppression mouse's pharmacodynamics experiment, rh-Tα1 can improve the ability of mouse's macrophage phagocytosis. The effective in the dose of 0.4 mg/kg and 0.8mg/kg are very obviouse(P<0.05). Rh-Tα1 can improve the ability of T lympholeukocyte transformation, the effective in the dose of 0.4 mg/kg and 0.8 mg/kg are obviouse(P<0.05). Rh-Tα1 can improve the ability of B lympholeukocyte transformation, the effective is obviouse in the dose of 0.2 mg/kg,0.4 mg/kg and 0.8mg/kg are aslo obviouse(P<0.05). Rh-Tα1 can raise the NK cell's activity in dose of 0.2mg/kg, 0.4mg/kg and 0.8mg/kg(P<0.05). It also can improve the level of IL-2. The effective are obviouse in the dose of 0.4mg/kg,0.8mg/kg (P<0.05);In the normal mouse's immunogenicity experiment, the dose of rh-Tα1 0.4mg/kg, 0.8mg/kg can obviously raise mouse's spleen index(P<0.01), the dose of 0.2mg/kg can can obviously raise thymus index. the dose of 0.2mg/kg can improve the ability of mouse's macrophage phagocytosis(P<0.01). rh-Tα1 can improve the ability of T lympholeukocyte transformation, the effective is obviouse in the dose of 0.2 mg/kg (P<0.05). Rh-Tα1 can improve the ability of B lympholeukocyte transformation, the effective is obviouse in the dose of 0.2 mg/kg,0.4 mg/kg and 0.8mg/kg(P<0.05). Rh-Tα1 can raise the NK cell's activity in dose of 0.2mg/kg, 0.4mg/kg and 0.8mg/kg. It also can improve the level of IL-2, the effective is obviouse in the dose of 0.8mg/kg(P<0.01).Conclusion:Rh-Tα1 has obviously pharmacodynamics effect of raising mouse's specificity immune function and non-specificity immune function in immumosuppression mouse. and normal mouse.
