Objective:To observe the effects of "Xindaxin" in treating depression model mice on brain and blood of active of SOD and content of MDA,NO,and to observe the state of immunization disorder on depression model mice and the effect of adjusting immunization and T lymphocyte o In order to provide the mechanism of "Xindaxin" anti-MD from improving the state of cell immune restrain and from cleaning serum LPO side,and provide a pharmacology foundation of "Xindaxin"for clinicalusage.Methods:1.The effect of "Xindaxin"on the behavior of depression model mice:(1)adopt CUMS that is currently extensive applicationby domestic and international,imitate to result in depression model mice.(2)Measure the mice weight and psychosis changes by week,measured the consumption of 1%sucrose solution every two weeks,and measured the mice behavior at the 28th and the 56th day.2.The effect of "Xindaxin"on brain and blood of active of SOD and content of MDA,NO of depression model mice:establish the depression model mice,study the effect of the drugs on brain and blood of active of SOD of depression model of by xanthine oxidase method.Study the effect of the drugs on brain and blood of content of MDA by NR method,establish the depression model mice on brain and blood of content of NO by TBA method.3.The effect of "Xindaxin" on the immune function of depression model mice: establish the depression model mice,Study the effect of the drugs on weight of the immune organ of depression model by detecting spleen index,thymus index.Study the effect of the drugs on thymusdependent lymphocytes subgroup of the peripheral blood of depression model by APAAP method.Results:1.The effect of"Xindaxin" on improving the behavior of depression model mice: compare with the normal group,every groups of mice been stimulated,the increment of weight number decrease obviously,the mental state and behavior all have changed.At the 28th day of experiment,by the method of"Bi an",every groups of mice been stimulated have more wrong times than normal group(P<0.05),the weight number is less than normal group(P<0.05),and the consumption of 1% sucrose solution is less than normal group(P<0.05) too.At the 56th day of experiment,compare with the normal group by using three dosage of Xindaxin and BaiYoujie group infusing stomach treatment,the medicine high and middle groups'weight number is more than normal group(P<0.05),and the medicine high and middle groups'weight number was superior to the model group(P<0.05),they have move times more than the normal group,and the medicine high groups'move times more than the model group(P<0.05),three dosage of Xindaxin have less wrong times than the normal group(P<0.05),and the medicine middle groups'was superior to the model group on wrong times(P<0.05),the consumption of 1% sucrosesolution is more than normal group(P<0.05).2.The effect of"Xindaxin" on raising the activity of serum SOD,and declining content of MDA,NO of depression model mice:compare with the normal group, the model group's activity of serum SOD and the serum NO,MDA levels the model group is lower than normal group(P<0.05),but the activity of serum SOD raised obviously,and the serum NO,MDA levels declined obviously in every treatment group(P<0.05),and three dosage of Xindaxin was not superior to BaiYoujie group.3.The effect of "Xindaxin"on enchaing the immune function of depression model mice:compare with the normal group,the model group's spleen index and thymus index were reduce very much(P<0.05).Its quantities of CD3,CD4 cell level of the peripheral blood in mice reduce and CD8 cell level increase,its CD4/CD8 ratio of the peripheral blood in mice was unbalance(P<0.05),but spleen index and thymus index were raised obviously(P<0.05 ),in three dosage of "Xindaxin",their quantities of CD3,CD4,CD8 cell level of the peripheral blood in mice were raised(P<0.05).Conclusion:1."Xindaxin" can resume the increment of weight number in the depression model mice,decrease the wrong times of mice in "Bi an" and increase the move times in"Zifa"experiment,increase the consumption of 1%sucrose solution and have the effect on improving the state of depression mice。Raised obviously activity of serum SOD and declined the serum NO,MDA levels in the depression model mice, the changes in LPO was possibility related to the mechanism of MD.The result of the experiment can prove the mechanism of Xindaxin anti-MD from serum LPO side and from clean O2R,and provide a pharmacology foundation of for "Xindaxin" clinical use.2 "Xindaxin" can increase spleen index and thymus index in the depression model mice,raise their quantities of CD3,CD4,CD8 cell level of the peripheral blood in mice.It can improve the state of cell immune restrain and have obvious corrective action to cell immune function in the depression model mice,the state of cell immune restrain was possibility related to the mechanism of MD.The result of the experiment can prove the mechanism of Xindaxin by improving the state of cell immune restrain, and provide a pharmacology foundation of Xindaxin for clinical use.
