ObjectiveStudy on the antidepressant Pharmacodynamics of Xindaxin compound,to investigate the levels of monoamine transmitters in pallium of mice,to elucidate the mechanism action of Xindaxin compound in immunological and endocrine function。Methods1.Pharmacodynamics research of Xindaxin compound1.1 Effect of Xindaxin compound on the forced swimming test(FST) with miceThe mice were divided into 6 groups,orally given for 7 days.30 minutes after the last drug period,mice were put to the instrument of forced swimming test;record the rotatory locomotors activities during forced swimming test.1.2 Effect of xindaxin compound on the chronic mild stress(CMS) depression model with miceAnimals were randomly separated into 6 groups.The CMS procedure employed here has been described previously,and was designed to maximize the unpredictable nature of the stressors.The CMS group was exposed to the following stressors in random order:water deprivation,food deprivation,water and food deprivation,damp bedding(300 mL water spilled into bedding),45°cage tilt,tail clamping for 1 minute, ice water swimming for 3 minutes.The stressors were presented in for a total of 4 weeks.Result1.Compared with the results of each group in the FST,we found that Xindaxin compound can increase the rotator locomotors activities during forced swimming test.2.After a total of 4 weeks unpredictable stresses,we found that the Xindaxin compound can increase the ambulatory activities,while the error number of entries to the dark compartment of rats in the step-through test were reduced.3.The mechanism action of Xindaxin compound in antidepressant may be followed:(1) CMS induced a decrease in the levels of NE and DA in pallium of mouse,while Xindaxin compound can improve it.(2) CMS induced endocrine disturbance of HPA axle,raise the levels of corticosterone in blood plasma and Xindaxin compound regulated it.(3) Plasma cytokines imbalance were existed in CMS,the Xindaxin compound could significantly promote the levels of IL-2 in blood plasma.
