| In this thesis,the fluorescence properties of Osthol were studied.The influence of experimental conditions on fluorescence spectroscopy was inspected.The fluorescence spectra of Frutus cnidii,Radix Angelicae Pubescentis,Murraya and Agrimoniae,as well as Chinese patent drugs Zhuanggu joint Pill and Zhonghua bonesetter Pill,were studied.New methods for analysis of Osthol in the Chinese medicines were established.The main contents of the thesis were as follows:1.The endogenous fluorescence and sensitized fluorescence properties of Osthol were studied.The impacts of acidity,organic solvents,ultraviolet radiation,β-cyclodextrin(β-CD) on fluorescence spectroscopy were inspected.It was found that Osthol itself was a weak fluorescent substance.Under ultraviolet radiation or inβ-CD solution,polymerization of it may occur so that its fluorescence intensity increased gradually.The impact of organic solvents on fluorescence intensity was remarkable.2.The fluorescence properties of Frutus cnidii were studied.It was found that the fluorescent properties Frutus cnidii was similar to Osthol.Fluorescence of Osthol in Frutus cnidii can be sensitized byβ-CD,while other components can not be sensitized.Using the sensitized fluorescence,a selective method for determination of Osthol in Frutus cnidii was established,and the result was 2.0%.The new method was verified by a thin-layer fluorescence scanning method and a HPLC-fluorescence detection method,the results of three methods were basically the same.3.The fluorescence spectra and HPLC map of Radix Angelicae Pubescentis were studied, and many fluorescent components,similar with Osthol,were found in Radix Angelicae Pubescentis.Due to the interference of these fluorescent components,the sensitization ofβ-CD on osthol couldn't be observed,so that Osthol in Radix Angelicae Pubescentis could not be determined byβ-CD-sensitized fluorimetric method.A HPLC-fluorescence detection method and a HPLC-absorption detection method were used to analyze Osthole in Radix Angelicae Pubescentis,the results obtained were 0.76%and 0.79%,respectively.4.HPLC-fluorescence detection method was used for analysis of Osthol in two Chinese medicines Murraya and Agrimony,but the chromatographic map showed that there was no detectable Osthol in these two drugs.5.The fluorescence spectra of Chinese patent drugs Zhuanggu joint pill and Zhonghua bonesetter pill were studied,and HPLC-fluorescence detection method and HPLC-absorption detection method were used respectively for determination of Osthole in two drugs,the obtained results were in substantial agreement. |
