| Fluorescence spectra of Podophyllotoxin and crude drugs containing Podophyllotoxin were studied in this thesis. Influences of experimental conditions on fluorescence spectra were also investigated. A thin-layer fluorescence scanning method was established for the determination of Podophyllotoxin in Podophyllum, Dysosma and Sinopodophyllum Ying (fruit). In addition, 3D fluorescence spectra of these drugs and their counterfeit drugs were studied, thereby a new approach for identifying these drugs was provided.Three parts were included in this thesis:1. Fluorescence spectra of Podophyllotoxin were studied. It was found that Podophyllotoxin present strong and steady fluorescence in acidic solutions with pH 2~6 and alkaline solutions with pH 11~13. Using L-tryptophan as a reference, fluorescence quantum yield of Podophyllotoxin was measured to be 0.12 under neutral conditions, and 0.14 under alkaline conditions. Fluorescence property of Podophyllotoxin on silica gel plate was studied, and a good linear relationship between fluorescence intensity and quality was found. Thereby a thin-layer fluorescence scanning method was proposed for the determination of Podophyllotoxin in crude drugs.2. Fluorescence spectra and thin-layer chromatogram of extracting solution of Podophyllum, Dysosma and Sinopodophyllum Ying (fruit) were studied, and several fluorescent components were found in these drugs. So, solution fluorescent method can not be used directly to analyze these drugs. Using thin-layer fluorescence scanning method, the content of Podophyllotoxin in Podophyllum was determined to be 1.50%, in Dysosma was 8.45%, and in Sinopodophyllum Ying (fruit) was 0.0245%.3. Three-dimensional fluorescence fingerprint spectra of Podophyllum, Dysosma, Sinopodophyllum Ying (fruit), Diphylleia sinensis Li and their counterfeit drugs were studied, and 3D fluorescence examination method for these drugs were established. |
