Objective:To explore the expression of CD117,C-erbB-2,NDRG1 in human hepatocellular carcinoma(HCC) and its significance;to provide the basis for molecular targeted therapy for the future,and for further analysis of the correlation of CD117,C-erbB-2 and NDRG1 in tumor's size,location,metastasis and histopathological classification,grading,staging,etc.METHODS:The expression of CD117,C-erbB-2 and NDRG1 were evaluated by EnVision immunohistochemistry staining in fetal liver tissues,normal liver tissues (liver transplant),cirrhosis liver tissues,transitional hepatocyte tissues, HCC,metastasis carcinoma tissues,and their average optical by image analyzer motic med 6.0.RESULTS:The average optical density of CD117,C-erbB-2 and NDRG1 in fetal liver tissues, normal liver tissues,cirrhosis liver tissues,transitional hepatocyte tissues,HCC,metastasis carcinoma tissues was respectively(0.177±0.130), (0.197±0.090),(0.159±0.086),(0.087±0.088),(0.069±0.088),(0.067±0.070);(0.031±0.034),(0.000±0.000),(0.001±0.001),(0.029±0.078), (0.028±0.065),(0.005±0.017);(0.059±0.074),(0.206±0.056),(0.198±0.074),(0.059±0.051),(0.128±0.096),(0.176±0.083).CD117,NDRG1 in fetal liver tissues,normal liver tissues,cirrhosis liver tissues,transitional hepatocyte tissues,HCC,metastasis carcinoma tissues of the average optical density gradually decreases,and the overall difference among the groups were statistically significant(P<0.05);C-erbB-2 in fetal liver tissues,normal liver tissues,cirrhosis liver tissues,transitional hepatocyte tissues,HCC, metastasis carcinoma tissues of the average optical density increase and the overall differences among the groups were statistically significant(P<0.05); but the expression of CD117 distinctly relates to the age,liver cirrhosis and PT(P<0.05);the expression of C-erbB-2 distinctly relates to the number of tumors,infection of HCV,peritoneal the visceral penetration(P<0.05); the expression of the NDRG1 distinctly relates to the Edmondson classification of carcinoma(P<0.05).CONCLUSION:CD117 involved in the occurrence and development of cancer,and could be thought as the HCC suppressor gene;C-erbB-2 could be thought as HCC cancer genes,promoting tumor occurrence and development;NDRG1 in HCC could be thought as cancer suppressor gene;there was a negative correlation between CD117 and C-erbB-2(r=-0.360,P<0.05);there was a significant negative correlation between C-erbB-2 and NDRG1(r=-0.669,P<0.05);there was a positively correlation between CD117 and NDRG1(r=0.377,P<0.05),there also was a synergistic effect between CD117 and NDRG1 in the inhibition of the occurence of HCC;the integration of CD117,C-erbB-2 and NDRG1 determine the prognosis of HCC.
