ObjectThrough assaying the plasma levels of adiponectin respectively in colorectal carcinoma patients and healthy controls and detecting adiponectin receptors R1 and R2 in colorectal carcinoma and normal surrounding cancerous discuss the relationship between plasma adiponectin,adiponectin receptors and colorectal carcinoma.MethodsWe collected plasma samples from 76 patients,including 38 colorectal cancer cases who was newly diagnosed colorectal cancer by pathohistologic confirmation of colorectal cancer at the Shanghai sixth People's Hospital and 38 healthy controls.Preoperative plasma samples were analyzed with a specific ELISA assay.We collected colorectal cancer tissue samples and paired neighboring noncancer colorectal tissue samples from 38 colorectal cancer patients and examined relative expression of adiponectin receptors AdipoR1 and AdipoR2 using immunohistochemistry.ResultsThe mean adiponectin levels in colorectal cancer patients and healthy controls were 159.89 and 184.5 pg/ml,respectively.Obviously plasma adiponectin concentrations of colorectal cancer patients were significantly lower than healthy controls(P=0.001).And a inverse correlation was found between plasma adiponectin levels and tumor size with significantly lower levels of adiponectin in tumors≥5cm(P=0.04),while no similar relationship was found between adiponectin and p53,Dukes stages(P>0.05) Adiponectin receptors,AdipoR1/AdipoR2,were expressed in neighboring noncancer colorectal tissue samples and colon cancer tissue samples.But expression of AdipoR1&R2 was found to be higher in tumor tissue compared with the patient's normal surrounding cancerous tissues(P=0.011,P=0.012).And expression of AdipoR2 was higher in tumor tissue which negatively express of the p53(P=0.018).Neither AdipoR1 nor AdipoR2 expression was significantly associated with the degree of tumor Size and Dukes stages.ConclusionsLower plasma levels of adiponectin are associated closely with colorectal carcinogenesis.And the Lower Plasma levels of diponectin were positively related to the degree of tumor Size.Both AdipoR1 and AdipoR2,were expressed in colorectal cancer tissue samples and neighboring noncancer colorectal tissue samples.Moreovre expression of AdipoR1&R2 was higher in tumor tissue compared with the normal surrounding cancerous tissues.We can presume that adiponectin and adiponectin receptors play a vital role in the process of colorectal carcinogenesis.Detecting adiponectin maybe a effective tool in the early diagnosis and prevent of colorectal carcinogenesis and also help to evaluate the curative effect of postoperation.Thus,there is a wide prospect of adiponectin and adiponectin receptors applying in clinical with other means for colorectal carcinogenesis.
