Abstract/Summary:
Objective:Oxidative stress is a hallmark of the diabetes state.In this study tempol (4-hydroxy-2,2,6,6-four methys piperidine),the ROS(reactive oxygen species) scavenger,which can penetrate the cell membrane of the ROS,was used to decreases oxidative stress and improves kidney function in diabetes rats.Methods:18 male SD rats(weight 200±25)g from Animal Center of Shanxi Medical University were used and were randomly divided into three groups,normal control(NC) group,diabetes mellitus(DM) group and diabetes add tempol(D + T) group to carry out this study.Diabetes rat models with streptozotocin(STZ)-induced were performed as describe.D + T group used every day by gavage given oral tempol,the other group given normal saline orally.4 weeks later,after 24h urinary albumin excretion rate(UAER) were examined,they were weighted,anesthetized and removed the kidneys.Malondialdehyde(MDA) content and superoxide dismutase(SOD), catalase(CAT) glutathione peroxidase(GSH-PX) activity in renal tissue were measured,too. They were perfused via the ascending aorta with saline followed by 500ml of 4%paraformaldehyde.Just removed kidneys,spinal cord,such as dehydration Organize fixed, frozen sections for NADPH oxidase subunit p22phox immunohistochemical staining.Results:24-hour urinary albumin(UA1b/24h),kidney weight / body weight(KW/BW) and malondialdehyde(MDA) content,superoxide dismutase(SOD) activity,glutathione -Px (GSH-Px) activity of DM group were significantly increased than those of NC group.However,there was no difference in catalase(CAT) activity,SOD,GSH-Px and CAT activity between D+T group and DM group.Immunohistochemical staining showed that within the renal parenchyma in renal tubular cell membrane protein expression for p22phox brown circle or lines,NC rare negative group, DM group showed diffuse distribution of a lot of strong positive,D+T group showed scattered weak positiveConclusion: In diabetic rats with tempol intervention,inhibition of reactive oxygen species ROS generation and p22phox to the kidneys to reduce the level of oxidative stress in the diabetic rats can delay the development of diabetic nephropathy.
