ObjectiveTo explore to choose a safe dose of NaHS,observe the impact with intravenous injection of hydrogen sulfide (H2S) donor sodium hydrosulfide (NaHS) in normal Wistar rats ,.MethodsRandom divided 32 the health, male and adult Wistar rats into control group:NaHS 2.8μmol/kg group; NaHS 8.4μmol/kg group; NaHS 14μmol/kg group; the hemodynamic changes were measured separately after intravenous administration at 0.5h, 1h, 2h, 4h (check four parameters of LVSP, LVEDP and±dp/dt max).After 4 h, the plasma H2S concentration was determined by methylene blue spectrophotometric method ,and the plasma TNF-α, MPO concentrations were measured by ELISA. At the same time, the heart, liver, brain, lung and kidney were stained by HE and observed the cell morphological changes by light microscopeResultsCompared with control group, there was no significant difference between NaHS 2.8μmol/kg, 8.4μmol/kg group, 14μmol/kg group at different moment about hemodynamic changes respectively (P>0.05).After administration 4h, the plasma H2S concentration increased significantly in NaHS 2.8μmol/kg (43.56±1.74μmol/L), 8.4μmol/kg (48.34±1.39μmol/L),14μmol/kg (57.25±1.72μmol/L) group compared with control group (39.26±1.83μmol/L) respectively(P<0.05).After administration 4h, the plasma TNF-αconcentration was no significant difference in NaHS 2.8μmol/kg (149.84±10.67 pg/ml), 8.4μmol/kg (157.27±10.16 pg/ml) group compared with control group (147.47±10.13 pg/ml) (P>0.05) and the plasma MPO concentration was no significant difference in NaHS 2.8μmol/kg (2.01±0.16 ng/ml), 8.4μmol/kg (2.08±0.12 ng/ml) group compared with control group(1.88±0.16 ng/ml) (P>0.05). the plasma TNF-α(172.48±11.18 pg/ml) and MPO(2.74±0.20 ng/ml) concentra -tions increased significantly in NaHS 14μmol/kg group compared with control, NaHS 2.8μmol/kg and 8.4μmol/kg group respectively (P<0.05).We found that cellularedema, inflamematory cell infiltration and hemorrhage in the rat lung tissue when injected into 14μmol/kg NaHS for 4 hour.Conclusion1 NaHS 14μmol/kg intravenous injection can increase the TNF-α, MPO concentration of the normal rats plasma and make morphological changes in rats lungs.2 It is safe to the rat body with the doses of 8.4μmol/kg NaHS and 2.8μmol/kg by intravenous injection. In order to observe the largest role in myocardial protection, the dosage will be selected 8.4μmol/kg.
