| Objective: To investigate the effect of exogenous sodium hydrosulfide on the secretion of TM,t-PA,PAI-1 and s ERCP by endothelial cells of burned rats,and to further clarify the protective effect of sodium hydrosulfide on endothelial cells.Methods: The research object of this experiment is the endothelial cells of SD rats.After subculture,the cells were treated to the same state and grouped by the end of the third generation.The composition of the intervention medium consisted of serum,treated with 150 μ mmol/L sodium hydrosulfide(Na HS)and 200 μ mmol/L glibenclamide(Gli).The experiment was divided into five groups: simulated burn group,burn group,burn+Gli group,burn+Na HS group,burn+Gli+Na HS group.Endothelial cells were treated by group.The expression levels of TM,t-PA and PAI-1m RNA in each group of cells were detected by q PCR technology;the expression level of s ERCP protein in each group of cells was detected by ELisa technology.Results :1.The expressions of TM,PAI-1 m RNA and s ERCP protein in each group were measured under different intervention conditions:(1)burn group>simulated burn group;(2)burn group> burn + Na HS group;(3)burn group +Gli group>burn group;(4)Burn+Gli group>burn+Gli+Na HS>burn+Na HS group.2.The results of measuring t-PA m RNA expression in each group:(1)burn group <simulated burn group;(2)burn group < burn + Na HS group;(3)burn group + Gli group < burn group;(4)burn + Gli Group<Burn+Gli+Na HS<Burn+Na HS group.The difference among the five groups was statistically significant,P<0.05.Conclusion : 1.Small amount of exogenous sodium hydrosulfide can reduce the damage of endothelial cells in burns.2.Small amount of exogenous sodium hydrosulfide can improve the function of the body’s coagulation system by affecting the secretion of endothelial cells intervened by burn serum. |
