Effect Of Caspase-1 Inhibitor On Acute Gouty Arthritis Induced By Monosodium Urate Crystals In Rats

OBJECTIVE:To study the effect of caspase-1 inhibitor on acute gouty arthritis induced by monosodium urate in rats.METHODS:Monosodium urate crystals were used to establish gouty arthritis model rats, Fifty male Wistar rats(180～220g) were randomized to five groups:the Controls(groupA), gouty arthritis group not be treated(groupB),Colchicine group(groupC),low AC-YVAD-CMK group(groupD),high AC-YVAD-CMK group(groupE),All the drugs were given for three days after modeling.Ankle joints's welling degree were measured by tape at different time(2h,4h,6h,8h,24h,48h,72h)after modeling.At 72h after modeling,all the rats were killed and the blood and joints rinse solution was collected for leucocyte count and IL-1βlevels were monitored by Radioimmunoassay.Pathological changes of articular synovium of rats were observed with HE staining.RESULTS:AC-YVAD-CMK could significantly inhibit joint swelling of rat and cut down leucocytes of blood and joints rinse solution(P＜0.01 );Radio-immunoassay revealed that AC-YVAD-CMK could reduce levels of IL-1βin rats'blood and joints rinse solution (P＜0.01).Histopathologically,AC-YVAD-CMK could abate synovium proliferation of ankle joint,reduce inflammatory cell infiltration of articlar tissues. CONCLUSION:At 72h after modeling,AC-YVAD-CMK have remarkable efficacy in improving symptoms of acute gouty arthritis induced by monosodium urate crystals in rats,which might be related to its inhibition on levels of IL-1β.
