Objective Through detecting the expression of MMP-3,MMP-7and TIMP-1 and DNA mutations of TIMP-1 in gastrointestinal stromal tumors(GISTs)to disscuss the relationship between their expressions and clincopathological characteristics and provide theoretical basis and probable indicators in order to predict metastasis potentiality of the tumor.Methods①The expression of MMP-3,MMP-7and TIMP-1 in 68 cases of GISTs were detected by immunohistochemical PV9000 method and SP method.②TIMP-1 gene mutations in exon2,exon 3 and exon4 were examined in these 68 specimens of GISTs using PCR-SSCP analyses,and the positive specimens were confirmed by DNA sequencing.③The immunohistochemical results were analyzed using the software of SPSS 13.0 by means of Rank sum test and Kendall's rank correlation analysis.Results①The expression(u=20.00,u=4.00,u=261.00,P<0.01)of MMP-3,MMP-7 and TIMP-1 in GISTs were higher than that of in normal gastric tissues.The result was of statistical significance.②The expression rate(X~2=9.547,P<0.01) and intensity(r=0.289,P<0.01)of MMP-3 had positive correlation with the pathological mitotic figures of GISTs.The expression rate and intensity(u=46.00,r=0.277,P<0.05)of MMP-7 had positive correlation with the metastasis of GISTs.③Significant correlation had been found between MMP-7 and TIMP-1(r=0.277,P<0.05)in GISTs.No significant correlation had been found between MMP-3 and MMP-7(r=0.191,P>0.05)in GISTs.④PCR-SSCP and silver staining displayed that TIMP-1 exon2,exon3 and exon4 in all the 68 GISTs had no mutations.Conclusions MMP-3,MMP-7 and TIMP-1 expressed negatively in normal gastric tissue,but intensively in GISTs.The difference was significant.The results indicated that the abnomal expression of MMP-3,MMP-7 and TIMP-1 were related with GISTs growth.The expression level of MMP-3 and MMP-7 were respectively positive-correlated with the pathological mitotic figures and metastasis of GISTs.These two may serve as markers to judge the clinical prognosis of GISTs.The expression of MMP-3 and MMP-7 in GISTs with different sizes showed no differences.We infered that size should not be used as criterion to judge GISTs.Significant correlation had been found between MMP-7 and TIMP-1 in GISTs.No significant correlation had been found between MMP-3 and MMP-7 in GISTs. We had not detected any proof to indicate correlation of the gene mutation and the higher expression of TIMP-1.We infered that the insufficinet compensation of MMPs/TIMPs leaded to high expression level of TIMP-1 in GISTs.
