Objectives: To study the relativity between P~16 genetic mutation and ovarian epithelial tumor by polymerase chain reaction—single stranded conformation polymorphism(PCR——SSCP). Materials and methods:1. 28 speciments of ovarian epithelial tumor that were tissue pieces blended with paraffin wax were collected .Of all these, 11 speciments were benign ovarian tumor and 4 speciments were neutral tumor,and 13 specciments were ovarian cancer . 2. P~16 genetic mutation was detected by PCR——SSCP tinting.Results:9 of 28 speciments of ovarian tumor were detected in alteration of form of mobile zone. This indicated that part of P~16 gene changed. The rate of the mutation was 53.57%(15/28).Of all these ,the rate of P~16 genetic alteration of the benign ovarian tumors was 72.7%(8/11)and that of the neutral ovarian tumors was 75%(3/4),and that of the ovarian cancers was 30.8%(4/13).Conclusion:1.Genetic mutation of ovarian epithelial tumors was found and P~16 genetic mutation perhaps promoted occurrence and development of ovarian tumor in the form of mutation .2. P~16 genetic mutation played a role in primary stage of ovarian epithelial tumor. The rate of P~16 genetic mutation varied in different stage of the tumor. 3.Technology of PCR—SSCP tinting was simple ,rapid and sensitive,and had not radio-active isotopes pollution. PCR—SSCP EB tinting was also widely used for testing genetic mutation,especially,for molecular biology of tumor .
