Objective To approach the effect of integrin-β1 antisense oligodeoxynucleotide(ASODN) on hepatic metastasis of human colon cancer(HT-29).Methods Integrin-β1 ASODN was transfected into HT-29 cell by liposome. The expression of integrin-β1 mRNA was determined by RT-PCR; Transfected HT-29 were injected into subcapsule of spleen in nude mice by open surgery.To establish a model of hepatic metastases of human colon cancer in nude mice and observe the effect of integrin-β1 ASODN on hepatic metastasis of HT-29.Results The expression level of integrin-β1 mRNA was significantly decreased in the ASODN group compared with that of non-sense oligodeoxynucleotide(NSODN) group and control group(P<0.01);the hepatic metastasis rate was 34.7%and 83.4%,the tumor inhibition rate was 73.02%and 5.25%,respectively.Conclusions ASODN decrease the expression level of integrin-β1 mRNA, and inhibit hepatic metastasis of HT-29 in nude mice.
